Research and development of new drugs is the core driving force in the field of medical development,but also the necessary force to promote social development.In recent years,with the development of systems biology and bioinformatics,the pharmaceutical industry has developed thousands of drugs for the treatment of diseases,of which a considerable number has been approved by the FDA.Collection of these drugs and their target information,not only contribute to the discovery of new drug targets,but also to promote the development of network pharmacology,contribute to multi-target drugs and combination of drug research and development.Kinases,as key molecules in intracellular signaling,play an important regulatory role in the physiological processes such as cell proliferation,differentiation,migration,and so on,which can lead to disease,especially cancer.So from the human kinase group to dig more new anti-cancer targets,and expand the field of disease from which to become the current development of new drugs as a hot spot.Based on the above problems,this topic first conducted a comprehensive and detailed study of drugs and targets,and integrated into the database TTD,and then based on network pharmacology on the targeted kinase drugs made a complete analysis.Detail steps of this study were listed as below:First,the upgrade of the "therapeutic utility drug target database" TTD,the upgrade content includes: 1)significantly increased the number of clinical trials of drugs and their targets.Increasing the number of clinical drugs and targets in the database to 9528 and 723,becoming the most comprehensive database of clinical drug and target information collection so far;updated 2071 listed drugs,17,803 preclinical study drugs and 397 responses Target,and further enhance the integrity and accuracy of the application of the database in the development of new drugs,2)the establishment of the TTD database of drugs,targets and KEGG,MetaCyc / BioCyc,Net Path,PANTHER pathway,PathWhiz,PID,Reactome And Wiki Pathways,etc.3)more convenient access to multi-target drug and combination of drug information.Second,based on network pharmacology,we studied the clinical targeting of kinases: the discovery of successful and clinical trials of kinase targets in the system level differences;reveal the human kinase group was multi-target drug effects and The difference between the kinases acting on the combination of drugs.Studies have shown that successful kinase targets tend to have fewer similar proteins outside their ownfamily of proteins,participate in the regulation of fewer biological pathways,and specifically distribute in fewer biological tissues.For kinase targets that are at different clinical stages Point of view,with the clinical changes in the state,but also show this trend;multi-target drugs are simultaneously targeted at the kinase family within a number of targets,and the combination of drug targets tend to kinase and non-kinase target Between the combination.
