Objective : Esophageal cancer is one of the most common human digestive tract malignant tumor which threat the health of human seriously,the exact pathogenesis is still unclear.It is widely believed that life style,driking,smoking and genetic reason were related to the pathopoiesis.Kiss-1 and PPAR? plays important roles in the development of a variety of malignant tumors of human beings.But the ralationship of esophageal cancer between the two genes was rarely reported.In the work,we examined the expression of Kiss-1 and PPAR? in tissues by qRT-PCR,and the relationship between genes and pathological files were analyzed,provide a theoretical foundation for understanding the role of Kiss-1 and PPAR? take part in the pathogenesis of esophageal cancer.Methods:1 40 cases fresh specimens were collected,included carcinoma tissues and normal mucous tissues.All the specimens were come from the First Hospital of Hebei Medical University and Fourth Hospital of Hebei Medical University between January,2012 and December 2015.All the patients were not received chemotherapy and radiotherapy before operation.2 The expression of Kiss-1 and PPAR? in 40 cases cancer tissues and correspond normal tissues were detected by qRT-PCR,and analyzed the relationship between the expression of Kiss-1 and PPAR? and clinicopathologic feature.3 All the results were analyzed by SPSS19.0 and GraphPad Prism 5.0,the results were showed by median(quartile),Wilcoxon test was used to analyzed paired samples,P<0.05 was regarded as statistical significance.Results:1 The expression of Kiss-1 in esophageal cancer tissues and normal tissues were 0.0256(0.0090,0.0965)and 0.0781(0.0424 0.3108),respectively.The expression of Kiss-1 in cancer tissues was lower than in normal tissues(P<0.05),the difference was statistically significance;2 The expression of Kiss-1 in lymph nodes metastasis were lower than no lymph nodes metastasis,the difference was statistically significance(P<0.01).Kiss-1 was independent with age,gender,depth of invasion,differentiated degree and TNM stage;3 The expression of PPAR? in esophageal cancer tissues and normal tissues were 0.2624(0.0677,0.5715)and 0.5280(0.1225 1.1839),respectively.The expression of PPAR? in cancer tissues was lower than in normal tissues(P<0.05),the difference was statistically significance;4 The expression of PPAR? in lymph nodes metastasis were lower than no lymph nodes metastasis(P<0.05),and the expression of PPAR? in low differentiative were lower than the high differentiation(P<0.01),the difference was statistically significance respectively.PPAR? was independent with age,gender,depth of invasion and TNM stage.Conclusions:1 The expression of Kiss-1 is low level in esophageal carcinoma,suggesting that Kiss-1 may probably have a potential tumor suppressor gene activity.2 The expression of Kiss-1 is down-regulated in the lymph nodes metastasis,suggesting that it has significance in judging metastasis malignancy and prognosis of esophageal carcinoma.3 The expression of PPAR? is low level in esophageal carcinoma,suggesting that PPAR?may probably have a potential tumor suppressor gene activity.The expression of PPAR? is down-regulated in the lymph nodes metastasis and low differentiative,suggesting that it has significance in judging metastasis malignancy and prognosis of esophageal carcinoma.
