| Objective To explore the influence of chronic unpredictable stress(UCMS)in SAMP8 mice on depression,learning and memory ability.Explore UCMS on brain derived neurotrophic factor(BDNF),tyrosine kinase receptor(Trk B)and neuron apoptosis related factor B cell lymphoma-2(Bcl-2),Bcl-2 Associated X Protein(Bax)protein expression.Methods Conduct chronic unpredictable stress on SAMP8 mice,then evaluate the ability of learning and memory through water maze test,and evaluate depression through sugar water preference test.Get the protein expression of BDNF,Trk B,Bcl-2and Bax in hippocampus through western blotting.Results 1 The incubation period of finding the platform increased significantly in stress group compared to control group(P< 0.05);the number of crossing platform in stress group after removing the platform decreased significantly compared to control group(P<0.05).2 Sugar water consumption of stress group decrased significantly compared to control group(P < 0.05).3 The protein expression of BDNF,Trk B in hippocampus of stress group decreased significantly compared to control group(P<0.05).4 The protein expression of Bcl-2 decreased and the protein expression of Bax increased in hippocampus of stree group significantly compared to control group(P<0.05).Conclusion The ability of leraning and memory became bad after stress.The protein expression of BDNF and Trk B decreased in hippocampus,the protein expression of Bcl-2 decreased and the protein expression of Bax protein increased in hippocampus. |
