ObjectiveTo explore the mechanism of..Kaiqiao-yizhi prescription ameliomicees the learning-memory ability of subacut aging mices, by observing the change of mices’behavior with Y-maze test and hippocampi’s micro-structure through microscope,examining the change of enzymoLogy in hippocampi with ultraviolet spectrophotometer.Methods78ICR which were4moon’s age were adjustably bred for1week,7mices fell into disuse for out of the90%region by morns water maze test,the rest71mices were divided into6groups:blank group(10mices), Positive control (11mices), Negative control(11mices), drug(low. middle, high)groups(13mices each). Day morning in the normal control group,0.3mL distilled water; the afternoon0.2mL distilled water and in0.2mL saline by intraperitoneal injection, continuous42days. Negative control group with0.3mL distilled water every day morning; pm25mg/mL intraperitoneal injection of D-gal (D-galactose)(200mg/kg)0.2mL, and to18.75mg/mL for AlCl3(150mg/kg)0.2mL gavage, continuous42days. Experimental group (low, medium and high-dose group) were fed daily morning0.1mL,0.2mL,0.3mL Kaiqiao-yizhi soup; pm intraperitoneal injection25mg/mL of D-gal(200mg/kg)0.2mL, and to18.75mg/mL of AlCl3(Aluminum chloride)(150mg/kg)0.2mL gavage, continuous42days. The morning of every day western medicine group0.03mg/mL of Huperzine A (0.4mg/kg)0.3mL gavage; PM intraperitoneal injection25mg/mL of D-gal(200mg/kg)0.2mL; and to18.75mg/mL of AlCl3(150mg/kg)0.2mL gavage, continuous42days. Experimental observation on the behavior of20min before intraperitoneal injection of scopolamine hydrobromide(SCOP)2mg/kg.The mices were texecuted after experiment, detecting the content of MDA and enzymoLogy activity of SOD(Superoxide dismutase).AchE in hippocampi of mices by ultraviolet spectro-photometer,observing the hippocampi’s micro-structure through microscope.ResultsAfter the experiment, Model control group of mice to complete the learning and memory required rate was significantly increased,signal discrimination accurately reflects the significantly reduced;brain weight was significantly decreased brain total protein content was significantly reduced brain tissue and serum MDA were significantly increased, SOD activity was significantly AchE activity decreased and increased; Pathology revealed:Fuzzy outline the hippocampus, neurons branch less condensed nucleoli smaller, segmented unclear, there is a broken away, can see a lot of swelling and degeneration of nerve cells, cells loose, blurring the lines between cells.The drug groups were better than model group in above of all targets after taked medicine,and related to the dose with direct miceio.Middle drug group was close to blank group,and had scarcely discrepancy eAch other, but the high group had greay discrepancy with the blank group.ConclusionKaiqiao-Yizhi prescription could promote the learning-memory ability of subacut aging mices. and the mechanism was below:First,D-gal,AlCl3and successfully induced by SCOP to replicate sets of aging and learning and memory in mice model.Second, Kaiqiao-Yizhi prescription subacute aging mice on learning and memory functions in promoting effects.Third,Kaiqiao-Yizhi prescription had the effect of anti-free radical.Fourth,Kaiqiao-Yizhi prescription can only reduce the formation of MDA.Fifth,Kaiqiao-Yizhi prescription could reduce the activity of AchE.Sixth,Kaiqiao-Yizhi prescription could lighten the damage of neuron when it took harm,and safeguarded the cell.
