Type 2 diabetes has become one of the most common chronic systemic diseases, the main performance of patients with persistent high blood sugar, sugar, fat, protein metabolism disorders. The incidence of the world is growing, causing serious medical, social and economic problems. Many studies have confirmed that oral disease and type 2 diabetes, the relationship between the close, such as: periodontal disease and so on.They play a mutual feedback between the role.In the process of its interaction, oral microbes are closely related to oral and systemic systemic health and disease. In this study,Illumina sequencing technique was used to study the diversity and diversity of oral flora in patients with type 2 diabetes mellitus and their normal subjects. And the difference in flora between oral saliva and gingival plaque in patients with type 2 diabetes mellitus. To study the correlation between glycosylated hemoglobin and fasting blood glucose in patients with type 2 diabetes mellitus through model correspondence analysis. The above study provides new clues for understanding the structure of oral flora in patients with type 2 diabetes mellitus and the relationship with systemic diseases. Objective: 1.To understand the diversity and diversity of oral flora in patients with type 2 diabetes mellitus and their normal subjects.2.To understand the difference of microbial flora between oral microflora of patients with type 2 diabetes mellitus.3.To understand the correlation between oral flora and some clinical indicators in patients with type 2 diabetes mellitus. Methods: 1.To investigate the diversity and composition of oral flora in patients with type 2 diabetes mellitus and non-diabetic patients. All the subjects selected from the Fourth Affiliated Hospital of the Fourth Military Medical University were included in the patients with type 2 diabetes and their relatives(with the patient as husband and wife). All subjects were divided into two groups. One group was type 2 diabetes and one was non-diabetic.Collected samples of saliva and gingival plaque were extracted from the sample. The data were compared with the RDP database to analyze the diversity and composition of oral flora in patients with type 2 diabetes mellitus and normal subjects, using the Illumina Hiseq PE250 platform for sequencing.2.To investigate the difference of oral salivary flora and gingival plaque in patients with type 2 diabetes mellitus. The investigate continued to use the method of experiment 1 to compare the oral microenvironment of patients with type 2 diabetes by comparing the composition of oral saliva and gingival plaque flora in patients with type 2 diabetes mellitus.3.To investigate the relationship between oral microflora and glycosylated hemoglobin and fasting blood glucose in patients with type 2 diabetes mellitus? In this study, the relationship between environmental factors, samples, and flora was examined by redundancy analysis(RDA)to explore the relationship between oral flora and systemic disease in patients with type 2 diabetes mellitus Relationship. Results: 1.In this study,18 phylum,33 Class,40 Order,80 Family and 153 Genus were found in 91 samples of oral flora. There was a significant difference in the comparison of oral flora between type 2 diabetes mellitus and normal subjects, but the difference was not significant.(P> 0.05).However,11 cases(P <0.05)were found at all levels of oral saliva and plaque in patients with type 2 diabetes mellitus and normal subjects in some significant differences in genotypes.2.There were significant differences in saliva and gingival plaque between type 2 diabetes mellitus and 39(P <0.05)at the level of salivary and gingival flora in patients with type 2 diabetes mellitus. And 95 species(P <0.05)were found at all levels with significant differences. The results showed that the microbial composition of oral microbes in patients with type 2 diabetes mellitus was different.3.In the analysis of the correlation between glycosylated hemoglobin and fasting blood glucose in patients with type 2 diabetes mellitus,we found that there were 30 species of genotypes that were positively correlated with glycosylated hemoglobin(P <0.05).There were 15 species of pathogens with fasting blood glucose(P <0.05).Conclusions: 1.There was no significant difference between the two groups of patients with type 2 diabetes mellitus and normal subjects.2.There were significant differences in the microbial composition of oral microbes in patients with type 2 diabetes mellitus under different microenvironment.3.Type 2 diabetes mellitus in some oral bacteria and patients with glycosylated hemoglobin, fasting blood glucose has a positive correlation. These flora may be associated with the development of oral diseases and systemic diseases.
