Objective:This study is aimed to determine the value of m RNA and protein expression level of two marker genes containing flavin 3 single oxidase(FMO3,high in the oviduct epithelial tissue expression),loss of brain malignant tumor gene(DMBT1,expressed in endometrial tissue)in ectopic endometrium tissue of ovarian endometriosis(EMs)with infertility and fertility patients and normal ovarian tissue.Methods:We studied patients with surgery treatment in Qing Dao University Inffiliated Hospital April 2014 and August 2015.The numble of 90 ovarian endometriosis patients is divided into 40 ovarian EMs+infertility patients and 50 ovarin EMs+pregnancy patients according to whether the patients accompany with infertility or not.The ectopic tissue is selected as observation group and control group respectively.The 35 cases of normal ovarian tissues from the Uterine fibroids surgery patients as normal group.The expression of FMO3,DMBT1 m RNA is detected by RT-PCR and protein is measured by Western Blot..Results: 1.The m RNA expression of FMO3,DMBT1 were significantly higher in observation group compared with normal group(P<0.05),with fold change were2.058-6.096(average 4.793)? 22.073-53.097(average 33.953)respectively.2.The m RNA expression DMBT1 was higher in the control group compared with normal group(P<0.05),with fold change was 5.892-12.436(average 9.619).The m RNA expression of FMO3 was not significantly different between control group and normal group(P>0.05),with fold change was 1.501-2.131(average 1.816).3.The m RNA expression of FMO3 were significantly higher in observation group compared with control group(P<0.05),with fold change was 1.738-8.500(average 3.847).The m RNA expression of DMBT1 was not significantly different between observation group and control group(P>0.05),with fold change was 1.013-4.514(average 2.116).4.The protein expression of DMBT1?FMO3 is 0.726-0.843(average 0.780)?0.659-0.723(average 0.699).The protein expression of FMO3,DMBT1 were significantly higher in observation group compared with normal group(P<0.05),with level were1.834-2.756(average 1.600)?1.143-1.356(average 1.259)respectively.5.The protein expression DMBT1 was higher in the control group compared with normal group(P<0.05),with level was 1.565-2.286(average 1.400).The protein expression of FMO3 was not significantly different between control group and normal group(P>0.05),with level was 0.826-0.941(average 0.880).6.The protein expression of FMO3 were significantly higher in observation group compared with control group(P<0.05).The protein expression of DMBT1 was not significantly different between observation group and control group(P>0.05).Conclusion:The ectopic lesions are highly expressed FMO3,DMBT1,in patients with ovarian endometriosis-associated infertility and is probably derived from both tubal epithelium and endometrial tissue;Ectopic lesions have only high DMBT1 expression level and the expression of FMO3 is no obvious rise ovarian EMs patients combined with children,prompting that endometrial tissue is the only source of ectopic lesions;The sources of tubal in ectopic tissue may be related with ovarian endometriosis-associated infertility.
