Blood analysis provides large amount of information for personal health,helping the professionals to determine the physiological status of various organs,so that they can diagnose corresponding disease.In the last decade,Quantitative Phase Microscopy(QPM)has been developed rapidly in classification of cells and morphology analysis due to its advantages such as non-invasive,rapid and efficient,and so on.Especially for the homogeneous red blood cell,Quantitative Phase Microscopy has made a breakthrough,success in achieving the morphological parameters of red blood cells,measuring on the chemical composition and dynamics study of red blood cells.However,due to the special structure of the heterogeneous cells,such as white blood cells and platelets,the application of the phase microscopy method for the detection of the heterogeneous cells remains to be developed.In this paper,digital holographic phase microscopy is introduced to illustrate the principle and characteristics of Interference phase microscopy.Quantitative phase microscopy based on the transport of intensity equation is introduced to illustrate phase microscopy under intensity analysis modulation.The optical path and the recovery method of three typical digital holography technology are emphatically analyzed in this paper.The stability,sampling speed and space bandwidth utilization rate of these three technologies are compared and analyzed as well.In this paper,the phase imaging software Biophase,based on the theory of transport of intensity equation,and Olympus GX51 were combined as phase microscopic imaging system.Respectively,red blood cells,platelets and neutrophils were texted by this phase microscopy imaging system.According to the optical characteristics of blood cell,the phase gradient analysis method and the main parameters of the routine analysis method applied to white blood cells,platelets and red blood cell,is put forward to characteristics analysis of blood cells.The method can be used to classify the blood cells into three or five groups.Researches,based on the sepsis platelet activation model,were carried out onplatelet using the imaging system above for the clinical study and dynamics analysis of platelet,under the cooperation with medical researchers.Other imaging technology(such as scanning electron microscopy)were also used to confirm the exogenous CO can inhibit platelet form releasing ? granule under the threat of LPS.The phase gradient analysis method and the main parameters of the routine analysis method,put forward in this paper,were applied to the analysis of platelets in different status,and achieve many useful results.Therefore,the value of the characteristics analysis method can be proved.We believe that those methods by further development can play a more important role in the field of blood examination.
