Cancer is one of the major threats to human health,and cervical cancer is the second largest malignant tumor to women.A large number of studies have shown that the effect of early cancer treatment is significantly better than the middle and advanced cancer.This means that if we could have an accurate diagnosis of cancer in the early stage,we could improve the life quality of patients and even improve the survival of patients with fixed number of year.For the early diagnosis of cancer,the main method is to detect the tumor markers by immune histochemistry method,molecular biology and flow cytometry.However,these methods have some shortcomings,such as false positive and false negative problems.Therefore,the label-free detection of cancer is becoming a researching spot.Atomic force microscopy has been widely used because of its characteristics of nanometer resolution and no damage to cells.In this paper,we used atomic force microscopy to detect the change of the characteristics of cervical cancer cells,and detect the pathological changes in the cells,and diagnose the cancer during the early stages.At the same time,in order to explore the treatment of cervical cancer,we further studied the characteristics of cerv ical cancer cells treated with docetaxel.Under the same measuring conditions,we used atomic force microscopy to detect the normal cervical epithelial cells and cervical cancer cell line HeLa before and after the treatment with docetaxel,compared the stress curves,and studied the changes of physical and chemical properties of cells.According to analysis the physical and chemical properties of cells after drug treatment,we have known physical and chemical properties have changed,cell plasticity index has changed,the internal structure of cell has changed,these changes have been used western blot assay to prove;the adhesion work has also changed,the modulus of elasticity of cells have changed,these changes have proved by transmission electron microscopy,through the transmission electron microscopy,we know microvilli on the surface of the cells treated with drug have significantly reduced;in addition,the elasticity of cells and others physical and chemical properties have also changed.Furthermore,the validity of the diagnosed result of atomic force microscopy was further verified by relevant biological experiments.The technology of atomic force microscopy used in cellular detection has made up the blank in this field,which makes it play an important role in the future scientific research.In addition,atomic force microscopy is also expected to become an important detection tool for medical detection.
