Research Purpose: Studying the protection of basil polysaccharide Polysaccharide to myelosuppression caused by cyclophosphamide can provide theoretical grounds for the application and further development of basil polysaccharide.Research Method 1: 50 healthy grownup Kunming mice which are allowed to acclimate for 3 days,the myelosuppression model of H22 tumor-bearing cell of which are copied.The mice are divided into 5 experimental groups: normal saline group,cyclophosphamide group(50mg/kg),group of basil polysaccharide 100mg/kg,group of basil polysaccharide 50mg/kg,and rh GCSF group.There are 10 mince in each group,to observe the effect of basil polysaccharide on survival rates of the experimental rice.Research Method 2: 60 healthy grownup Kunming mice which are allowed to acclimate for 3 days,the myelosuppression model of H22 tumor-bearing cell of which are copied.The mice are divided into 6 experimental groups: normal contrast group,normal saline group,cyclophosphamide group,group of basil polysaccharide 100mg/kg,group of basil polysaccharide 50mg/kg,and rh G-CSF group.There are 10 mince in each group,to observe their food-intake,to measure the number of their blood cells and morrow nucleated cells,tumor inhibition rates,spleen index,thymus index,and to make femoral section and PI dyeing to measure the marrow cell cycle by flow cytometry.Research Results: 1.The cyclophosphamide group,the two groups of basil polysaccharide,and rh G-CSF group all can boost the survival days and survival rates of the rice,and prolong the medial death time and medium survival time.Compared with the cyclophosphamide group,the group of basil polysaccharide 100mg/kg significantly prolongs the average survival days,survival rates,medial death time and medium survival time of the cyclophosphamide chemotherapy H22 tumor-bearing mice(P<0.05).The rh G-CSF group does not significantly affect the average survival days,survival rates,medial death time and medium survival time of the model mice.2.Since the 4th day of inoculation,there appear nodes at the right subaxillary where the inoculation is performed,size of a rice grain.With the growth of the tumor,the rice begins to be less glossy in fur,motionless,and less active in food-intake,especially the cyclophosphamide group.The average food-intakes(g)are as follows: normal contrast group:7.6±0.82,normal saline group:6.6±1.39,the cyclophosphamide group:5.2±1.03,the group of basil polysaccharide 100mg/kg:7.2±1.53,the group of basil polysaccharide 50mg/kg:6.2±1.09,and the rh G-CSF group:5.8±1.35.Compared with the normal contrast group,the food-intake of normal saline group is reduced significantly(P<0.05);Compared with the normal contrast group,the food-intake of cyclophosphamide group is reduced significantly(P<0.05);while compared with the cyclophosphamide group,the food-intake of normal saline group,the two groups of basil polysaccharide,the rh G-CSF group is increased significantly,especially the group of basil polysaccharide 100mg/kg being the most significantly increased group. 3.The number of white cells(×109/L): normal contrast group is 9.0±2.24,the normal saline group4.5±1.78,the cyclophosphamide group 2.1±0.64,the group of basil polysaccharide 100mg/kg 5.7±1.64,the rh GCSF group4.1±1.26.Compared with the normal contrast group,the number of white cells of the normal saline group decreases significantly(P<0.05)and compared with the normal saline group,the number of white cells decreases significantly(P<0.05),which means the modeling is a success.Compared with the cyclophosphamide group,the number of white cells of the group of basil polysaccharide 100mg/kg and the rh G-CSF group increases significantly(P<0.05),among which the number of white cells of the group of basil polysaccharide 100mg/kg is superior to the number of white cells of the rh G-CSF group.4.Tumor inhibition rate:the cyclophosphamide group is 64.67%,the group of basil polysaccharide 100mg/kg is 65.67%,the group of basil polysaccharide 50mg/kg is 48.76%,and the rh G-CSF group is 60.69%.Compared with the cyclophosphamide group,the tumor inhibition rate of the group of basil polysaccharide 100mg/kg is slightly higher.5.The spleen index and thymus index(mg/g):those indexes of the normal contrast group are 4.81±0.620 and 1.21±0.207;those indexes of the normal saline group are 3.23±0.532 and 0.92±0.143;those of the cyclophosphamide group are 2.14±0.312 and 0.41±0.078;those of the group of basil polysaccharide 100mg/kg are 4.53±0.617 and 0.80±0.178;those of the group of basil polysaccharide 50mg/kg are3.84±0.416and0.63±0.108,and those of the rh G-CSF group are 4.02±0.526 and 0.67±0.132.Compared with the normal saline group,the spleen index and the thymus index of the normal contrast group decrease significantly(P<0.05);compared with the normal saline group,the spleen index and the thymus index of the cyclophosphamide group decrease significantly(P<0.05),and compared with the cyclophosphamide group,the spleen index and the thymus index of the two groups of basil polysaccharide and the rh G-CSF group increase significantly(P<0.05),among which the spleen index and the thymus index of the group of basil polysaccharide 100mg/kg are significantly higher than the other groups.6.The number of morrow nucleated cells(×105/L):that of the normal contrast group is 178.4±8.29,that of the normal saline group is 102.4±18.4,that of the cyclophosphamide group is 57.8±15.23,the group of basil polysaccharide 100mg/kg is 153.4±15.63,that of the group of basil polysaccharide 50mg/kg is 153.4±15.63,and that of the rh G-CSF group is 70±41.7.Compared with the normal contrast group,the number of morrow nucleated cells of the normal saline group decreases significantly(P<0.05);compared with the normal saline group,the number of morrow nucleated cells of the cyclophosphamide group decreases significantly(P<0.05);and compared with the cyclophosphamide group,the number of morrow nucleated cells of the group of basil polysaccharide 100mg/kg increases significantly(P<0.05),among which the number of morrow nucleated cells of the group of basil polysaccharide 100mg/kg is superior to the group of basil polysaccharide 50mg/kg and the rh G-CSF group.7.In the femoral section of the mice,in the bone marrow cavity of the mice of the normal contrast group can see different lineages of blood cells in different development stages,in even distribution,having a few adipose cells and clearly visible microvessels and blood sinuses.In the normal saline group,different lineages of blood cells in different development stages are observable,in a slightly larger amount,active in growing,having a few adipose cells,and the blood sinuses are expanding and congesting.In the cyclophosphamide group,the blood cells of different lineages decrease in the number respectively,and the number of macrophagocytic cells decreases obviously,and megakaryocyte and Nucleated Red Blood Cells(NRBC)are still observable.In the group of basil polysaccharide 100mg/kg,the blood cells of different lineages decrease to a slight degree,the number of adipose cells increases,microvessels are expanding and the blood sinuses are congesting.In the group of basil polysaccharide 50mg/kg,the blood cells of different lineages decrease in the number respectively,the number of adipose cells becomes larger,microvessels are expanding and the blood sinuses are congesting.In the rh G-CSF group,the blood cells of different lineages recover basically,megakaryocyte slightly increases and the blood sinuses are expanding and congesting.8.Bone marrow cell cycle: Compared with the normal saline group,the G0/G1 period proportion of bone marrow cells of the mice in the normal contrast group rises significantly(P<0.05),S period proportion lowers significantly(P<0.05),and the proliferation index lowers significantly(P<0.05);compared with the normal saline group,the G0/G1 period proportion of bone marrow cells of the mice in the cyclophosphamide group rises significantly(P<0.05),G2/M period proportion rises significantly(P<0.05),the proliferation index lowers significantly(P<0.05);compared with the cyclophosphamide group,the G0/G1 period proportions of bone marrow cells of the mice in the group of basil polysaccharide and the rh G-CSF group lowers significantly(P<0.05),S period proportion rises significantly(P<0.05)the proliferation index rises significantly(P<0.05),among which the proliferation index of the group of basil polysaccharide100mg/kg rises most significantly.Conclusion: The basil polysaccharide 100mg/kg can significantly improve the survival rates,food-intake,the number of white cells,the number of morrow nucleated cells,the tumor inhibition rate,the spleen index,the thymus index and the incremental index of the morrow cells of H22 tumor-bearing mice which have undertaken cyclophosphamide chemotherapy.The histological section of the thighbone indicates that basil polysaccharide 100mg/kg can decrease the reduction of all types of blood cells because of the cyclophosphamide chemotherapy.It shows that basil polysaccharide can significantly protect the myelosuppression of the mice caused by cyclophosphamide chemotherapy.Moderating the cell apoptosis and enhancing the multiplication of the marrow cells might be one of the molecular mechanisms that basil polysaccharide can protect the myelosuppression of the tumor-bearing mice caused by cyclophosphamide chemotherapy.
