Objective: To study the effects of the basil polysaccharide to paclitaxel in vitro inhibition of human lung adenocarcinoma A549 cell line chemotherapy synergistic action and pharmacology and molecular mechanisms of action.Methods: MTT assay basil polysaccharide on inhibition of paclitaxel A549 cell proliferation, A549 by inverted phase contrast microscope morphological changes caused basil polysaccharide in combination with paclitaxel, and to analyze A549 cell death basil polysaccharide and paclitaxel caused by the use of Hoechst 33342/PI double staining method. Cell cycle was analyzed with Flow cytometry method after A549 cells stained with Propidium iodide. Confocal laser scanning microscopy and flow cytometry were utilized to detect mitochondrial membrane potential and concentration of calcium ion A549 cells which were stained with JC-1 and Fluo-3/AM seperately. Real-time quantitative PCR detected GST-?, MDR1 resistance group, and Bcl-2 and Bax expression of apoptotic gene.Results: MTT results and morphological observation table basil polysaccharide polysaccharide paclitaxel inhibition A549 cells have obvious synergies, the synergistic effect was concentration and time dependent; Hoechst 33342/PI Taxol and paclitaxel plus basil polysaccharide for 48 h apoptotic cells. Compared with paclitaxel treatment, basil polysaccharide in combination with paclitaxel drugs treatment could fibroblast cell cycle of G2/M cycle and S cycle and increase apoptotic cells; basil polysaccharide in combination with paclitaxel drugs treatment could decrease the intensity of red fluorescence, while green fluorescence was significantly enhanced, and mitochondrial membrane potential decreased;basil polysaccharide in combination with paclitaxel drugs treatment could significantly enhance the red fluorescence intensity, showing the elevated concentration of calcium ion in cytokinesis. Real-time quantitative PCR assay showed basil polysaccharide used in combination with paclitaxel significantly reduced MDR1, GST-? expression and down regulated Bcl-2, regulate the expression of Bax.Conclusion: Basil polysaccharide having chemotherapy synergies, can promote paclitaxel inhibition of human non-small cell carcinoma A549 cells proliferation in concentration and time dependent, and to induce apoptosis as the main mode. Basil polysaccharide in combination with paclitaxel drug treatment could increase mitochondrial membrane permeability, elevate concentration of calcium ion in cytokinesis and decrease mitochondrial membrane potential which affected the cell cycle, And basil polysaccharide in combination with paclitaxel reduced by Bcl-2, regulated Bax, thereby inducing apoptosis, increased apoptosis; down MDR1 and GST-?, thereby reducing tumor cell resistance to chemotherapy and improve the intracellular concentration of the drug; to achieve enhanced paclitaxel inhibit tumor growth, to strengthen the synergistic effect of chemotherapy.
