Objective: Based on follicular fluid metabolomics analysis of “four seven” and “five seven” female follicular fluid,to explore the mechanism of oocyte senescence,identify and determine the age of infertile kidney qi deficiency syndrome are closely related to the difference between the scientific prediction and Diagnosis of old age infertility kidney qi deficiency syndrome,to explore the nature of its syndromes to do a preliminary attempt to enrich the level of follicular fluid from the level of “Nei Jing” “seven seven” theory of scientific connotation.Methods: A retrospective analysis of 55 patients into IVF/ICSI cycles infertility patients,screening criteria according to age will be divided into two groups: age group(35 to 42 years old,27 cases,“five seven”)and the young group(28 to 35 years old,28 cases,“four seven”),choose plan of antagonists super stimulate ovulation,on two groups of patients was egg number,maturation,fertilization rate,cleavage rate,high quality embryo rate,clinical pregnancy rate and abortion rate,live birth rate were analyzed.Experimental study using ultra high performance liquid chromatography(HPLC)-high resolution mass spectrometry analysis of two groups of follicular fluid,to obtain multidimensional chromatography data using principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)processing,will be differences between the groups of compounds database information matching,charge ratio,formula is finally obtained by fragment ions scanning pieces of information on the structures of these substances to determine differences in compounds.Results:Clinical research: The older group and younger group based on age,sinus follicle has a very significant difference(P<0.01),FSH levels had significant difference(P<0.05),with statistical significance.In a number of eggs,MII egg number,the number of normal fertilized egg,the effective number of embryos have extremely significant difference(P<0.01),group-was lower than that in young group,with statistical significance.High quality embryo number,abnormal fertilized egg number has no obvious difference(P>0.05),no statistical significance.In the normal fertilization rate had significant difference(P<0.05),group-was lower than that in young group,with statistical significance.In mature,abnormal fertilization rate,cleavage rate,high quality embryo rate,cycle cancellation rate has no statistical significance(P>0.05).In the clinical pregnancy rate were extremely significant differences(P<0.01),group-was lower than that in young group,with statistical significance.In live births rate,miscarriage rate has no statistical significance(P>0.05).Experimental study: the difference between the two groups of compound roughly divided into hormonal protein,lecithin,lysophosphatide and degradation of section 4 classes,in which there are 7 kinds of raised objects: 4,5-Dihydroorotic acid,Maleylacetoacetic acid,4-oxo-Retinoic acid,Rhazidigenine Nb-oxide,Deoxycorticosterone,3-hydroxynonanoyl carnitine,5,6-Dihydrouri-dine,cut material has six kinds: DG(14:1(9Z)/22: 2(13Z,16Z)/0:0)? LysoPC(14:0)? LysoPC(16:0)? LysoPC(18:0)?TG(18:1(11Z)/24:0/20:5(5Z,8Z,11 Z,14Z,17Z))?Phytosphingosine,only the differences of compounds found in follicular fluid in the older group has two kinds: Nicotine glucuronide,Phosphaticlyl-choline(16:0/22:0).Conclusions: 1.The old and young group compared to the obvious reproductive function decline,reveals the connotation of TCM theory of "groups".2.Found that the differences of compound in the DAG(14:1(9Z)/22:2(13Z,16Z)/0:0)? TAG(18:1(11Z)/ 24:0/20:5(5Z,8Z,11 Z,14Z,17Z))? LysoPC(14:0)? LysoPC(16:0)? LysoPC(18:0)?Phosphaticlylcholine(16:0/22:0)?4-oxo-Retinoic acid?Nicotine glucuronide?3-hydroxynon-anoyl carnitine are closely related to the female reproductive function,affect the growth of female oocyte,mature,aging and the decay of the female reproductive function,speculated that the age for women at the beginning of kidney deficiency type screen.3.The differences between two groups of compounds did not look up to 4,5-Dihydroorotic acid?Maleylacetoacetic acid ? Rhazidigenine Nb-oxide ? Deoxycorticosterone ?3-hydroxynonanoyl carnitine?5,6-Dihydrouridine research data related to the female reproductive function,looked at in future studies to further explore its correlation.
