Objective: In order to explore the pathogenesis of phlegm dampness syndrome of PCOS from the aspect of metabonomics,and search the related biomarkers,the detection technology of ultra performance liquid chromatography-mass spectrometry(UPLC-MS)was employed to investigate the difference of follicular fluid metabolites between polycystic ovary syndrome(PCOS)patients and healthy people with vitro fertilization-embryo transfer(IVF-ET).The results will provide basis for clinical diagnosis and treatment of PCOS.Methods: Twenty patients with phlegm-dampness type PCOS and tweenty healthy follicles with IVF / ICSI were selected.The forty follicular fluid samples were analyzed by high performance liquid chromatography-high resolution mass spectrometry.After collecting all the data of follicular fluid samples,the method of principal component analysis(PCA)and partial least squares-discriminant analysis(PLS-DA)were used to analyze the data and get the metabolic fingerprinting.Multivariate statistical analysis of integral quantity was used to construct the score plot and analyze the deference between the two groups.And then choose the biomarkers of phlegm dampness syndrome of PCOS according to the analysis of different metabolic components.result: There were 17 differentially expressed compounds between PCOS group and normal healthy group.Comparing with the level of follicular fluid metabolites of the healthy people,the level of oleic acid,eicosatrienoic acid,eicosapentaenoic acid,docosapentaenoic acid,3-hydroxynonanoyl carnitine,dehydroepiandrosterone sulfate,deoxidized cortisol(14: 1(9Z)/ 22: 2(13Z,16Z)/ 0: 0)are higher in PCOS patients with phlegm dampness syndrome with statistically significant difference(P<0.05).However,the level of diastereomeric acid,(14Z),LysoPC(14: 0),LysoPC(16: 1),LysoPC(18: 0)are lower in PCOS patients,with statistically significant difference(P<0.05).Conclusion: Metabonomics can distinguish the healthy people and PCOS patients with phlegm dampness syndrome,and the biological markers of phlegm dampness syndrome of PCOS may be oleic acid,eicosatrienoic acid,eicosapentaenoic acid,docosahexaenoic acid,docosahexaenoic acid,(14: 0),LysoPC(16: 0),LysoPC(18: 0),3-hydroxynonanoyl carnitine,dehydroepiandrosterone sulfate,deoxycorticosterone,phenylalanine(18Z),24: 0/20: 5(5Z,8Z,11 Z,14Z,17Z)),DG(14: 1(9Z)/ 22: 2(13Z,16Z)/ 0: 0).The changes of follicular fluid metabolites in PCOS patients with phlegm dampness may be related to lipid metabolism,amino acid metabolism and hormone metabolism.
