Objective Statistics and summarized the clinical manifestations of PCOS patients with yang deficiency type,the use of data mining methods to analyze symptoms and characteristics of patients with kidney yang deficiency PCOS.Combined with follicular fluid metabonomics,to explore follicular fluid metabolic markers in patients with PCOS.From the perspectives of molecular biology and TCM syndromes,we studied the patients with PCOS with deficiency of kidney yang.Methods 1.Obtain the symptom data of PCOS patients through the clinical questionnaire,and use the data mining method to conduct statistical analysis.A total of 567 PCOS patients met the inclusion criteria,including 252 patients with PCOS with deficiency of kidney and315 patients with non-deficiency of PCOS.A total of 39 symptoms were collected.Univariate analysis,multivariate Logistic regression analysis and ROC curve analysis were used to analyze the distribution of common symptom characteristics in patients with PCOS and to explore the diagnostic criteria of PCOS.2.A retrospective analysis of 100 study-included PCOS patients who had undergone in-vitro fertilization was conducted and grouped according to the diagnostic criteria of TCM and Western medicine.Among them,33 cases of PCOS patients with kidney-yang deficiency type were set as observation group;67 cases of IVF-ET group were treated with simple male factor,and were set as control group.Taking the follicular fluid obtained from oviparous day as the research object,the method of principal component analysis(PCA)and Partial Least Squares-Discriminant Analysis(PLS-DA)were used to analyze the samples in an intuitive,reliable and statistically significant manner The difference of follicular fluid metabolites between the two groups was compared.Results 1.Kidney Yang type PCOS diagnosis of meaningful symptoms:aversion to cold,waist and knee pain,limbs cold,hi warm hi press,loss of libido,pale tongue.2.Kidney Yang deficiency PCOS follicular fluid metabolic differences include: 1.Leucine and isoleucine degradation pathway substances:2-Oxoisovalerate,6-Oxohexanoate,2-Oxoglutarate;2.Tyrosine metabolism pathway substances: L-Phenylalanine;3.Fatty acid metabolic pathways substances: Linoleate,Oleic acid,Docosahexaenoic acid;4.Glycosphingolipid metabolic pathways substances: Hexadecanal;5.Lipid metabolic pathway substances: Arachidonate,Lyso PC Lyso PC 18: 1,Lyso PC18: 0,Lyso PC 20: 5,Lyso PC 20: 4;6.Retinoic acid metabolism pathways substances: 13,14-Dihydroretinol;7.Cholic acid biosynthesis pathway substance: Lithocholic acid,Hyodeoxycholate,27-Hydroxycholesterol,3alpha,7alpha,12alpha-Trihydroxy-5beta-cholestan-26-al,5beta-cholestane-3alpha,7alpha,12 alpha,23,25-pentol.8.Vitamin metabolic pathway substances: Vitamin D3,25-Hydroxyvitamin D3,13'-hydroxy-alpha-tocopherol.9.Carnitine shuttle system pathway material: stearoylcarnitine.10.Cholesterol biosynthesis pathway substance: 5-Dehydroepisterol.Tryptophan pathway material: Tryptophan.Conclusion Data mining summarizes the clinical symptoms of kidney Yang deficiency PCOS patients,including aversion to cold,waist and knee pain,extremities cold,hi-hi press,loss of libido,pale tongue.For the diagnosis of kidney yang PCOS patients provide a reference standard.Follicular Fluid Metabolomics Analysis Compared follicular fluid metabolites in patients with kidney yang deficiency PCOS and control group were helpful to determine the biomarkers of PCOS in yang deficiency type,and then to explore its etiology and mechanism from the molecular biology point of view,But also provided a new way of thinking for its treatment.
