Objective:Construction of this study vocal damage in rabbit animal model,at different times after injury were injected into DCN,the use of HE and immunohistochemistry method,monitoring of TGF-?1 expression and distribution of variation.To observe whether the core proteoglycan has an inhibitory effect on scarring and to explore the mechanism of core proteoglycan inhibiting scar formation.Methods:25 New Zealand rabbits were used,5 rabbits were randomly selected as non-intervention group,20 rabbits in acute bilateral vocal cord injury models do.6 rabbits injected 100?g/L decorin immediately on the right vocal cord injury site,whereas the left vocal cord vocal cord injury site as a control.6 rabbits 1 months after injury on the right vocal cord injection of 100?g/L decorin site of injury,and the left vocal cord vocal cord injury site as a control.Growth in rabbits after a good,no infection,laryngeal edema.And surgical control group were 20 rabbits after 4 week and 12 week after injection harvesting vocal folds,Using HE and immunohistochemistry methord,detection of TGF-?1expression and distribution of variation,and then compared with values obtained in each group and statistically analyzed.Results:1.By the way of intercricothyrotomy injury of vocal cord tissue of the New Zealand rabbits,the animal model of vocal cord injury was successfully completed,it laid a foundation for the further study of whether exogenous injection of DCN could inhibit scar hyperplasia in vocal cords.2.The results of HE staining.The normal vocal cord HE staining showed the clear structure of the vocal cords,the epithelial layer,the lamina propria and the muscular layer,and the lamina propria was loose connective tissue.In the injury group,the fibrous tissue of the lamina propria of the vocal cords was increased,the collagen fibers were dense,the staining was deeper than that of the normal vocal cords,and the arrangement of collagen fibers was more disordered.In the injection group,the fiber structure of the vocal fold lamina was more than that of the normal vocal cords.2.The expressions of TGF-?1 by inmunohistochemical and it,s relationship with cinicopathologic characteristic in the experimental group and control group.(1)A,B group compared with the C group TGF-?1 expression high,there was statistical significance.(P <0.01).(2)A,B group on the right side of the vocal cords compared with the left vocal cords,TGF-?1 expression was significantly reduced,the difference was statistically significant(P <0.01).(3)Compared with A and B group,TGF-?1 expression was significantly reduced,the difference was statistically significant(P <0.01).(4)Compared with the two observation group of A and B,the TGF-?1 expression did not change significantly the difference was not statistically significant(p=0.17?p=0.14,p>0.05)Conclusion1.DCN can be highly compatible with collagen fibers in order to regulate the formation of collagen fibers,interfere with the stability of collagen fibers in the tissue,thereby changing the collagen fiber assembly and structure,thereby preventing scar formation.2.TGF-?1 is a key factor in the pathogenesis,DCN can inhibit the activity of TGF-?1 and inhibit the formation of scar by combining with TGF-?1.Scar formation in the early stage of fibrosis is relatively light,early injection of DCN effect is better,and DCN degradation rate is appropriate,is a good material for the prevention and treatment of scar.
