ObjectiveTo investigate the photo protective effects and the molecular transduction mechanisms of grape seed proanthocyanidin extract(GSPE)on HaCaT cells induced by UVA irradiation,we detected the expression of AQP3,Caspase 14 and BH,which are the key factors of the skin barrier.To reveal the photo protective function and the role of decreasing vascular hyperplasia of GSPE in early light aging,we detected the expression of VEGF and PEDF.We expect that GSPE can be used in medical skin care products to protect skin from the irradiation of ultraviolet.MethodsThere are three groups.HaCaT cells induced by UVA irradiation were pure light group.HaCaT cells induced by UVA irradiation and then cultured with GSPE at various concentration were GSPE group,and the third group is control group.The cells proliferation were detected by MTT method.To assess mRNA expressions of AQP3,Caspase 14,BH,VEGF and PEDF in different groups of HaCaT cells by RT-PCR method.The expression of VEGF was detected by ELISA method.And the expression of PEDF was detected by immunocytochemistry method.Results1.The impact on HaCaT cells proliferation is not obvious when the concentration of GSPE is lower than 200 μg/mL.And the 25 J/cm2 dose of UVA can inhibit the proliferation of HaCaT cells.2.The mRNA of AQP3 is increased in HaCaT cells induced by UVA irradiation.And that can be decreased gradually by GSPE.The mRNA of Caspase 14 and BH is decreased in HaCaT cells induced by UVA irradiation.And that can be increased gradually by GSPE of different concentrations in a dose dependent manner.The mRNA of AQP3,Caspase 14 and BH can be back to normal levels finally.3.The mRNA of VEGF is increased in HaCaT cells induced by UVA irradiation.And the secretion of VEGF protein is also increased.All of that can be decreased significantly by GSPE of different concentrations.And be back to normal level finally.The mRNA of PEDF is decreased in HaCaT cells induced by UVA irradiation.And the expression of VEGF protein is also decreased.And all of that can be increased gradually by GSPE in concentration-dependent manner.ConclusionsThe mRNA expression of AQP3,Caspase 14 and BH in HaCaT cells induced by UVA irradiation can be adjusted by GSPE.So that skin can maintain nomal hydration.And ensure that the physiological synthesis of natural moisturizing factor.Thus repair the skin barrier function that destroyed by UVA.Still GSPE can adjust the mRNA and protein expression of VEGF and PEDF in HaCaT cells induced by UVA irradiation.So the drug can reduce the formation of new blood vessels.Thus it can delay the light aging of our skin.
