Influence Of BDNF And ? Interferon On Astrocyte And Oligodendroglia And Inflammatory Factorsof EAE Rats

Objective To study the effect of brain derived neurotrophic factor(BDNF)on the growth of astrocyte and oligodendroglia in normal SD rats and experimental autoimmune encephalomyelitis(EAE)model,as well,the influence of BDNF and ?interferon on immune factors of EAE rats,to open up new ideas and provide some theoretical basis for the treatment of multiple sclerosis(sclerosis multiple,MS).Methods Through incomplete Freund's adjuvant and Mycobacterium tuberculosis to induce EAE animal model,to culture astrocyte and oligodendroglia of healthy control group and experimental group of EAE neonatal rat,after subculture,BDNF(0 ng/ml?0.1ng/ml?1ng/ml?10ng/ml)and ?interferon(0 ug/ml?0.01ug/ml?0.1ug/ml?1ug/ml)were added to the culture medium.Observation the change of the number of the astrocyte and oligodendroglia by immunofluorescence(mean±standard deviation),detect the level of anti-inflammatory cytokine interleukin-10(interleukin,-10,IL-10)and chemokine(The chemokine C-X-C motif ligand 1,CXCL1),as well as proinflammatory cytokine interleukin-1(interleukin-1,IL-1)and tumor necrosis factor alpha(Tumor Necrosis alpha factor-a,TNF-?)based on OD450 value analysisby enzyme-linked immunosorbent assay(ELISA)and statistical analysis.ResultsIn the experimental group and control group,the number of astrocyte and oligodendroglia increased after the addition of a certain concentration of BDNF(0 ng/ml?0.1 ng/m?1ng/ml?10ng/ml),and the number of astrocyte and oligodendroglia increased with the increase of concentration generally(mean±standard deviation).The study also found that BDNF(0 ng/ml?0.1ng/ml?1ng/ml?10ng/ml)and ? interferon(0 ug/ml?0.01ug/ml?0.1ug/ml?1ug/ml)can increase anti-inflammatory IL-10 and CXCL1 level(P<0.001),and reduce pro inflammatory cytokine IL-1 and TNF-a levels(P<0.001)in EAE SD rats of.Conclusion The study shows that BDNF can promote the growth of astrocyte and oligodendroglia,BDNF and ? interferon have anti-inflammatory effects,and may be valuable for MS.
