Objective:To study the correlation between the abnormal copy numbers of 5 genes(Prox-1 TERC,LAMP3,RYBP and GBE-1)and the prognosis of early cervical cancer.And to analyze the relationship between the abnormal expression of 5 genes and their clinicopathological features,investigating the prognostic significance of these markers in early cervical cancer.Methods:Screening the cases with postoperative pathological diagnosis of squamous cell carcinoma and clinical stage of early cervical cancer(stage ? a-? b)in 2008-2010(included 40 cases).Based on the results of follow-up visit,these cases were divided into 2 groups,the recurrence and metastatic group(12 cases)and the control group(28 cases).Collect paraffin fixed tumor tissue specimens of 40 cases,using DirLys-buffer method to extract DNA.Then operate the MLPA experiment(multiplex ligation dependent probe amplification)on the extracted tissue,to detect the copy number expression of gene Prox-1,TERC,LAMP3,RYBP and GBE-1.The gene expression levels between two groups were compared to analyze the correlation between the abnormal expression of these 5 genes and the prognosis and clinicopathological features of early cervical cancer.Results:Gene Prox-1,TERC and LAMP3 were amplified in early cervical cancer,while gene RYBP and GBE-1 were deleted.In the recurrence and metastasis group,the amplification expression rate of Prox-1 was 33.3%,the amplification expression rate of TERC and LAMP3 were 66.7%,RYBP and GBE-1 deletion expression rate was 33.3%,the gene copy number changes in the control group were 3.6%,39.3%,39.3%,10.7%,32.1%separately,which were lower than in recurrence and metastasis group.Only Prox-1 had shown a statistical correlation with prognosis(P=0.022).In association with clinicopathological features,there was a correlation between TERC and LAMP3 co-amplification and vascular lymph invasion(P=0.024).In the survival curve analysis,the 5 year disease-free survival rate(DFS)of patients with abnormal expression of Prox-1,TERC,LAMP3 and RYBP were significantly decreased.Conclusion:We found that the expression of Prox-1 was significantly correlated with the prognosis of early cervical cancer,suggesting that it was an independent prognostic marker.The abnormal expression of the other 4 genes did not show a statistically significant correlation with the prognosis.The consideration may be related to the small sample size.Further study with an expanded sample size is needed.
