Objective:To investigate the expression of Interferon regulatory factor 2 binding proteins 2(IRF2BP2)in gastric cancer and its role in proliferation and invasion,find a new target for the treatment of GC.Methods:1.Western blotting was used to assay the expression of IRF2BP2 in GC tissues and cells;Immunofluorescence was employed to detect the expression of IRF2BP2 locating GC cells;2.Adopted immunohistochemical staining method to detect the expression of IRF2BP2 in 65 cases of GC tissue specimens and 20 cases of noncancerous tissues;then analysed the relation of IRF2BP2 expression level with clinical pathological features and overall survival of GC.3.Using small interfering RNA(siRNA)to down-regulate the expression level of IRF2BP2 in GC cells(SGC-7901 and BGC-823),then divided the cells into 4 groups,which were negative contorl group,interfereing group(IRF2BP2 siRNA-1,IRF2BP2 siRNA-2,IRF2BP2 siRNA-3),respectively;the transfection efficiency was deteced by Western blot after transfection.4.The MTT assay and colony formation assay were conducted to evaluate the ability of proliferation of GC cells after transfection;5.Transwell assay was used to investigate the ability of migration and invasion of GC cells after transfection;Western blotting was used to assay the expression of EMT proteins after transfection.Results:1.The data of Western blotting show that IRF2BP2 was markedly overexpressed both in GC cells and tissues;Immunofluorescence showed that IRF2BP2 was localized in the nucleus;2.The result of immunohistochemistry found that the percentage of samples with a positive expression rate of IRF2BP2 protein in GC tissues was significantly greater than in the noncancerous(NC)tissues(P<0.001);The expression of IRF2BP2 significantly correlated with age,tumor size,clinical stage,depth of invasion,histological grade,and nerve invasion;The 5-year overall survival of the IRF2BP2 high expression group was significantly lower than that of the low expression group(P=0.0057);3.The proliferation and colony formation ability of GC cells was decreased after down-regulated IRF2BP2 expression;4.The migration and invasion ability of GC cells were decreased after down-regulated IRF2BP2 expression;The detection of EMT protein by Western blot showed that the expression of E-cadherin increased and the expression of N-cadherin and Vimentin decreased.Conclusion:1.IRF2BP2 was overexpressed in GC cells and tissues,and its expression was correlated with the invasive phenotype and poor prognosis of GC;2.In GC cells,knockdown IRF2BP2 can inhibit the ability of proliferation;3.In GC cells,knockdown IRF2BP2 can inhibit both migration and invasion ability and EMT.
