Objective The gold standard to diagnose Endometrial cancer is histopathology. when patients appeare irregular vaginal bleeding during postmenopausal,doctors will do diagnostic curettage to confirm the disease, depending on the patients' circumstances.But the diagnostic curettage is an invasive procedures, so many scholars committed to find some simpler, non-invasive way to diagnose endometrial adenocarcinoma.Known that the tumor markers Ca125 many relate with tumorigenesis, diagnosis, prognosis, and other aspects. but its accuracy and sensitivity is not high.Many studies have found that tumorigenesis relate with DNA damage repair. RAD51 (DNA duplexes repair genes) and BRCA1 (Breast cancer susceptibility gene 1) play a key role in DNA damage repair homologous recombination,and both aberrant gene expression give increasing attention in the process of tumor development. This experiment will explore the expression level of RAD51, BRCA1 in endometrial adenocarcinoma and their correlation, and both the relationship of histological grade, FIGO clinical stage, myometrial invasion, lymph node metastasis, the patient's age and other aspects in endometrial adenocarcinoma. Both are expected to become a new method for screening and diagnosis, new targets for treatment and new indicators of prognosis in endometrial cancer.Methods Collected 52 cases of endometrial adenocarcinoma and 45 cases of normal proliferative endometrium.Grouped as follows:age: 6 patients are younger than 45 years old,33 patients'age are between 46 with 60,13 patients are older than 61 years old; histological grade: 32 cases of well differentiated, 20 cases of poorly differentiated; FIGO clinical stage: 40 cases of ?-?,12 cases of ?-?;myometrial invasion: the depth of myometrial invasion in 39 patients is less than 1/2, the depth of myometrial invasion in 13 patients is greater than or equal 1/2; lymph node metastasis: 4 cases have lymph node metastasis, 48 cases without lymph node metastasis;Generally all subjects have no significant difference in national, living area, Underlying disease,et al. Immunohistochemistry was used to detect the expression levels of RAD51, BRCA1.Application SPSS19.0 software to statistical analysis,use chi-square test,rank sum test to analysis the expression both in endometrial carcinoma and normal endometrium tissues. Analyzing the correlation in endometrial adenocarcinoma with varying degrees of differentiation, FIGO stage,age, myometrial invasion, lymph node metastasis,et al.Results Immunohistochemistry showed that the positive rate of BRCA1(36.5%) was lower in endometrail adenocarcinoma than in proliferative endometrium(95.6%),and the difference have statistically significant (P<0.05).The positive rate of RAD51(80.8%) was higher in endometrail adenocarcinoma than in proliferative endometrium(15.6%), and the difference have statistically significant (P<0.05) (P<0.05). Both the expressions have no relationship of histological grade,FIGO clinical stage, myometrial invasion, lymph node metastasis, the patient's age and other aspects in endometrial adenocarcinoma,that is the difference have no statistically significant (P<0.05).Conclusion BRCA1 is lowly expressed in endometrail adenocarcinoma;RAD51 is highly expressed in endometrail adenocarcinoma;and both the expressions have no relationship of histological grade, FIGO clinical stage, myometrial invasion, lymph node metastasis, the patient's age and other aspects in endometrial adenocarcinoma. This experiment can preliminary prove that the expression of RAD51, BRCA1 are related with the occurrence and development of endometrial adenocarcinoma.
