ObjectiveTo detect the expression of C-X-C chemokine ligand-14(CXCL14)and Epidermal growth factor receptor(EGFR)in gastric cancer and non gastric cancer gastric mucosa tissues,including autologous and allogeneic non gastric cancer gastric mucosal tissues.To analyze the effect of CXCL14 and EGFR expression in the development and progression of gastric cancer,and the correlation between CXCL14 and EGFR.To provid the idea and reference for further related experimental research about the effect of CXCL14 in gastric cancer or other malignancies,and experimental basis about early detection,prognosis judgment or reliable molecular treatment targets of gastric cancer.Methods1.Specimen collection121 cases of postoperative gastric cancer specimens(Because the histological type of gastric cancer is mainly adenocarcinoma,so all the gastric cancer specimens in this experiment are gastric adenocarcinoma,that is expressed in gastric cancer in the paper),62 cases of autologous non gastric cancer gastric mucosa and 60 cases of allogeneic non gastric cancer gastric mucosa specimens were collected from 2015 to 2016.The diagnosis was confirmed by paraffin section and hematoxylin-eosin staining(HE),then immunohistochemistry(IHC)staining.At the same time,fresh specimens were collected from the above for HE,IHC staining specimens,including 41 cases of gastric cancer and 41 cases of autologous non gastric cancer gastric mucosa specimens,and immediately were loaded into the marked frozen tube and stored in liquid nitrogen at-176°C for m RNA detection.The patients corresponding to all specimens before surgery did not receive chemotherapy,radiotherapy and immunotherapy or others.The study was approved by the Ethics Committee of the Court.2.Experimental methodThe expression of CXCL14 and EGFR protein in 121 cases of gastric cancer,62 cases of autologous non gastric cancer gastric mucosa and 60 cases of allogeneic non gastric cancer gastric mucosa specimens were detected by immunohistochemistry staining.Relative expression of CXCL14 and EGFR m RNA in 41 cases of gastric cancer and 41 cases of autologous non gastric cancer gastric mucosa specimens were detected real-time quantitative RT-PCR.3.Statistical analysisSoftware SPSS 17.0 was used for above the data of each group analysis.The measurement data were presented in the form of mean and standard deviation(means±SD),then analyzed by paired t test.The count data were analyzed by ?2test.The correlation of CXCL14 and EGFR was analyzed by Spearman method.The significance level was set as P<0.05.Results1.The expression level of CXCL14The positive expression rate of CXCL14 protein in gastric cancer group,autologous non gastric cancer gastric mucosa and allogeneic non gastric cancer gastric mucosa specimens group were80.17%,61.29%,51.67%,respectively.There was significant difference between the three groups(P <0.01).The expression of CXCL14 protein in gastric cancer group was significantly higher than that in autologous non-gastric gastric mucosa group and allogeneic non gastric cancer gastric mucosa group(P<0.01)There was no significant difference between autologous group and allogeneic non gastric cancer gastric mucosa group(P>0.05).The expression of CXCL14 protein were correlated with the depth of invasion,differentiation and clinical TNM stage(P<0.05).The expression of CXCL14 protein was significantly higher in the serous group than in the non-serous group(P < 0.05).The patients with poorly differentiated gastric cancer were higher than those in the high-middle differentiation group(P <0.05).The clinical TNM stage ? + ? stage was higher than the ? + ? group(P <0.05).The relative expression of CXCL14 m RNA in gastric cancer group and autologous non-gastric gastric mucosa group were2.821±0.229,1.000±0.000.The differences were significant(P<0.01).The expression of CXCL14 m RNA were correlated with the depth of invasion,differentiation and clinical TNM stage(P<0.05).The relative expression of CXCL14 m RNA was higher in the serous group than in the non-serous group(P< 0.05).The patients with poorly differentiated gastric cancer were higher than those in the high-middle differentiation group(P <0.001).The clinical TNM stage? + ? stage was higher than the ? + ? group(P <0.05).2.The expression level of EGFRThe positive expression rate of EGFR protein in gastric cancer group,adjacent normal gastric mucosa group and allogeneic normal gastric mucosa group were 48.76%,30.64%,21.67%,respectively(P<0.01).The expression of CXCL14 protein in gastric cancer group was higher than that in non-gastric gastric mucosa group and allogeneic non gastric cancer gastric mucosa group(P<0.01).There was no significant difference between autologous group andallogeneic non gastric cancer gastric mucosa group(P>0.05).The expression of EGFR protein were correlated with the differentiation of gastric cancer,lymph node metastasis and clinical TNM stage(P<0.05).The patients with poorly differentiated gastric cancer were higher than those in the high-middle differentiation group(P <0.01).The presence of lymph node metastases was higher in patients without lymph node metastasis(P <0.05).The clinical TNM stage ? + ? stage was higher than the ? + ? group(P <0.01).The relative expression of EGFR m RNA in gastric cancer group was significantly higher than that in normal gastric mucosa group(P<0.01),which were(2.117±0.750,1.000±0.000),respectively.The expression of EGFR m RNA were significantly correlated with the differentiation of gastric cancer,lymph node metastasis and clinical TNM stage(P<0.05).The patients with poorly differentiated gastric cancer were higher than those in the high-middle differentiation group(P <0.001).The presence of lymph node metastases was higher in patients without lymph node metastasis(P <0.05).The clinical TNM stage ? + ? stage was higher than the ? + ? group(P <0.05).3.The correlation of CXCL14 and EGFRBased on Spearman Correlation analysis,the expression of CXCL14 protein in gastric cancer was positively correlated with the expression of EGFR protein(r = 0.195,P< 0.05).The relative expression level of CXCL14 m RNA in gastric cancer was positively correlated with the relative expression level of EGFR m RNA(r = 0.408,P< 0.01).Conclusions1.The expression of CXCL14 protein and m RNA in gastric cancer tissues was higher than that in non gastric cancer gastric mucosa tissues,and the expression of CXCL14 is related to the depth of invasion,differentiation and clinical TNM stage of gastric cancer.It suggests that CXCL14 may be an earlymolecular event,and expression of CXCL14 is involved in the development,progression and poor prognosis of gastric cancer.2.EGFR protein and m RNA expression in gastric cancer tissues were higher than those in non gastric cancer gastric mucosa tissues.Meanwhile,the expression of EGFR was correlated with the differentiation of gastric cancer,lymph node metastasis and clinical TNM stage.It suggests EGFR is involved in the development,progression and poor prognosis of gastric cancer.3.There is a positive correlation between the expression of CXCL14 and the expression of EGFR in gastric cancer tissues at the protein and gene levels,suggesting that they have certain synergistic effect in the development and progression of gastric cancer.Combined detection of CXCL14,EGFR may be more reference for the early diagnosis,the prognosis judement of gastric cancer.
