Preliminary Study On The Effects Of Astragalus Membranaceus Iniections On Nasopharyngeal Carcinoma

Abstract/Summary:
Objective: To investigate the effect of astragalus membranaceus injections on the biological behavior of nasopharyngeal carcinoma CNE-1 cells.Methods: After treated with different concentrations of astragalus membranaceus injections,the cell growth curve was determined by CCK-8 and the morphological changes of cells were observed by light microscope.The cell cycle and apoptosis of the nasopharyngeal carcinoma CNE-1 cells were detected by flow cytometry.The expression of caspase-3 was tested by western blot.The cell migration and invasion capabilities were tested by wound healing assay,transwell migration assay and transwell matrigel invasion assay,respectively.The adhesion ability was detected by cells-matrix adhesion experiment.Results:The results of CCK-8 assay showed that astragalus membranaceus injections could greatly inhibit the proliferation of human nasopharyngeal carcinoma CNE-1 cells,and the difference compared with the control group was statistically significant(P < 0.05).Under the microscope,the cell proliferation was significantly inhibited in the drug group,and the growth was slow.Theresults of flow cytometry showed that the astragalus membranaceus injections groups have less S phase cells(%)and more G0/G1 phase cells(%),higher apotosis rate(%)and more expression of caspase-3 than the control group(P<0.05).The wound healing assay indicated that the migration rate(%)was much lower than the control group(P<0.01)both in the first 12 hour and in the second12 hour.In the trans-well migration assay and trans-well matrigel invasion assay,the cells passed the trans-well membrane was less in the astragalus membranaceus injections group than the control group(P < 0.05).But no significant differences were found in the adhesion assay.Conclusion : The astragalus membranaceus injections can decrease the proliferation,migration abilities,invasion abilities and promote the apoptosis capability of nasopharyngeal carcinoma CNE-1 cells,in the same time,the cell cycle test shows that more cells were blocked in G1/S phase.
