To discuss the changes of endothelin and nitrix oxide levels in blood and liver tissue after the treatment of astragalus membranaceus injections and the protective effect of astragalus membranaceus injections in liver injury caused by experimental obstructive jaundice in rats. Methods: forty-six male SD rats were randomly divided into 4 groups: Normal control group(NC n=8), Sham operation group(SO n=8), Obstructive jaundice group(OJ n=15), Astragalus membranaceus injections group(AMI n=15). Models of Obstructive jaundice were made on rats by ligating and cutting common bile duct. After the bile duct ligation, Obstructive jaundice group were treated by physiological saline, while Astragalus membranaceus injections group were treated by Astragalus membranaceus injections. The levels of ET and NO in blood and liver tissue, the levels of ALT and TBi in blood were assayed on 14 day after the bile duct ligation. Meanwhile, the histopathological changes of liver tissue were observed.Results: the level changes of ET in blood and liver tissue had statistical significance after the bile duct ligation(F=30.24, P<0.01; F=293.92 P<0.01). The levels of ET in SO group had no statistical significance, compared with NC group(P>0.05). The levels of ET in OJ group increased significantly, compared with NC group and SO group (P<0.01). the level changes of NO in blood and liver tissue had statistical significance after the bile duct ligation(F=64.2 P<0.01;F=565.17 P<0.01). The levels of NO in SO group had no statistical significance, compared with NC group(P>0.05). The levels of NO in OJ group increased significantly, compared with NC group and SO group (P<0.01). The levels of ALT and TBi in blood serum increased significantly compared with NC group and SO group (P<0.01), just as the level changes of ET and NO. The levels of ET and NO in blood and liver tissue decreased significantly after the treatment of Astragalus Membranaceus Injections(P<0.01), but the it didn't decreased into normal level. The levels of ALT and TBi in blood serum decreased significantly in AMI group (P<0.01), but the it didn't decreased into normal level. Histological examination showed that liver cells were damaged remarkably in OJ group. The damages of liver cells abate in AMI group, compared with OJ group. NC group and SO group had no changes. Conclusion: From this study, we can drove these conclusion: 1 The levels of ET and NO in blood and liver tissue increased significantly after the bile duct ligation, compared with NC group and SO group. Furthermore, the levels of ALT and TBi in liver tissue also increased significantly. All these changes same as the histological changes. 2 From the treatment of Astragalus Membranaceus Injections, the levels of ET and NO in blood and liver tissue decreased significantly, liver function and liver cell damage abate. It suggested that AMI can effectively decrease the levels of ET and NO in blood and liver tissue after OJ and alleviate the damage of liver. AMI has protective effect on liver.
