Objective : To investigate the effects of Astragalus Membranaceus combined with Salvia Miltionhiza on tumor growth and metastasis, and to make an preliminary study of the involved biological mechanismas.Method: 1. In vivo experiments of tumor inhibitition: H22 hepatoma cells and S180 sarcomas cells were inoculated in mice to make models of tumor growth, and their difference in the general status, tumor weight, spleen and thymus weight were observed; the morphologic changes of tumor tissues were presented by HE staining and the levels of VEGF, ICAM-1 , VCAM-1,CD4 ahd CD8 were shown by the immunohistochemical method. 2. In vitro experiment (1) the effects of rat serum derived from Astragalus Membranaceus administration combined with Salvia Miltiorrhiza on the proliferation of human lung adenocarcinoma A549 cells were examined by inverted phase contrast microscope and MTT colorimetric analysis; (2) Labelling the A549 tumor dells with fluorescence to observe the effects of serum derived from Astragalus Membranaceus administration combined with Salvia Miltiorrhiza on the adhesion between A549 cells and human umbilical vein endothelial cells (HUVEC).Results: The tumor in Chinese medical treatment group weighed significantly less than those in the control ; In contrast to the control, the expression of VEGF,ICAM-1 and VCAM-1 diminished, and the expression of CD4 inceased significantly in tumor tissues. The experiments in vitro showed that serum derived from Astragalus Membranaceus administration combined with Salvia Miltiorrhiza could inhibit the proliferation of A549 cells and decrease the adhesion between A549 cells and HUVEC cells.Conclusions: Astragalus Membranaceus administration combined with Salvia Miltiorrhiza can suppress the metastasis tumors. The mechanism is related inhibiting the proliferation of tumor cells, decreasing the adhesion, and improving the immunological function of the body.
