Objective: To observe the protection effect of Xiaobanxia Decoction(XBXT)on gastrointestinal mucosal barrier in a chemotherapy rat model and to investigate the mechanism of XBXT in treating Gastrointestinal mucositis(GIM).Methods: Male Wistar rats were randomly divided into normal control group,XBXT normal control group,cisplatin model group,omeprazole group,and XBXT low,medium and high dose group.In addition to the first two control groups i.p.0.9% NaCl(10 mL/kg),other groups i.p.6 mg/kg cisplatin to establish chemotherapy rat model.Each group were given distilled water(10 mL/kg),0.8 g/kg XBXT,distilled water(10 mL/kg),omeprazole 1.2 mg/kg,0.4 g/kg XBXT,0.8 g/kg XBXT,1.6 g/kg XBXT in two days before i.p.cisplatin.Observed and recorded the rats' body weight,food intake and water intake at 24 h.After modeling 24,72 h selected 8 rats in each group and collected the blood,gastric antrum and ileum.Hematoxylin and eosin(HE)staining was used to observe the changes of gastric antrum and ileum,ELISA kit was used to measure the levels of MUC2,MUC5 ac and D-Lactate in serum,and immunohistochemistry was used to measure the levels of MUC2 in ileum,MUC5 ac in antrum and the Ki-67 in gastrointestinal mucosal cells.Results: In model rats,body weight and food intake was significantly lower than the normal animal,the levels of MUC2,MUC5 ac decreased,D-lactate increased in serum,the cell proliferation rate of gastrointestinal mucosa and the mucin expression was decreased(P<0.05).XBXT can effectively improve the body weight and food intake of rats induced by cisplatin,and increase the mucin in serum and tissue,and increase the cell proliferation rate(P<0.05).Conclusion: XBXT can inhibit the gastrointestinal mucositis,the mechanism of XBXT may be related to the protection in gastrointestinal mucosal barrier.
