Objective:The aim of this study is to investigate the protective effect and mechanism of external biliary drainage to Severe acute pancreatitis(SAP)model by 4% Sodium taurocholate solution was injected by retrograde puncture of pancreatic duct,and observe the expression of pancreas NF-?B P65 protein,the levels of serum HO-1,HMGB-1,TNF-? and IL-10 and pancreatic tissue pathology change.Methods:108 healthy female Sprague wrote-Dawley(SD)rats,4% sodium taurocholate solution was injected by retrograde puncture of pancreatic duct through duodenum in order to set up SAP model.The experimental animals were divided into control Group and model Group were:(1)Control Group not Drainage were divided into control Group(Sham Operation Group,SOG Group)and control Drainage Group(Sham Drainage Group,SDG Group),rats were randomly divided into 3 groups with 18 rats in each group.(2)Drainage of model Group was divided into not(Severe Acute Pancreatitis,SAP Group),the Drainage of model(Bile Drainage Group,BDG Group),rats were randomly divided into 3 groups with 18 rats in each group.(3)After the success of the groups in modeling of 3,6,12 h to rats(n=6)at each time point again after anesthesia laparotomy observation,and to take blood and pancreatic tissue,such as detection of AMY's index,using enzyme-linked immunoassay(ELISA)to detect serum cytokine TNF alpha,HO-1,HMGB1 and IL-10 expression and content,the protein immunoblot method(Western Blot)to determine the NF-kappa B protein expression in the pancreas.By HE staining in the rat pancreatic tissue observe its pathological changes.Results:After modeling,3h,6h,12h: The level of the serum AMY,TNF-a of BDG group were lower than SAP group(P<0.05).The level of the serum HMGB1 of BDG group is lower to the SAP group,(P<0.05).the serum IL-10 level of BDG group were compared with the SAP group is no statistical significance(P>0.05).The HO-1 serum level of BDG group were compared with SAP group were significantly higher(P<0.05).The level of the of the pancreatic NF-?BmRNA of BDG group is lower to SAP group in protein,there is significant difference(P<0.05)at3h,6h.The pathological grading value of BDG group compared with SAP group were significantly lower(P<0.05),and with the passage of time,the pathologic grading value showed a trend of increase in rats.Conclusion:(1)Improving the retrograde pancreatic puncture SAP rats model is a modeling method,which have simple operation,good repeatability,and certain stability,and it can meet bile drainage of pancreatic function protection research animal experiment needs;(2)To inhibit the body's release of proinflammatory cytokines and alleviate the pancreatic tissue damage,through improving anti-inflammatory cytokine expressionof of SAP outside bile drainage model rats.(3)The external biliary drainage can improve SAP the pancreatic tissue in rat models of inflammation of pathological changes and reduce pancreatic tissue damage.
