Objective:To discuss the relationship between expression of serum miRNA-181a and OSAHS patients,by measuring the serum miRNA-181a in OSAHS patients of different types and analyzing the correlation related to PSG index.Methods:1.Collect the clinical date of 41 cases male OSAHS patients with polysomnography(PSG)between September 2016 and January 2016 in Head and Neck Sleep Apnea Center of the First Affiliated Hospital of Kunming Medical University.All the paitents are aged 3 5?65.According to the PSG results of OSAHS in Chinese Medical Association clinic guidelines to divide into two groups:20cases of mild OSAHS group and 21 cases of medium and severe OSAHS group.At the same time examined 19 healthy people as a control group.Collect the basic information and PSG results of the subjects.All subjects excluded chronic pulmonary disease,hypertension and other cardiovascular and cerebrovascular diseases,diabetes and other endocrine disorders,thyroid disease,nutritional metabolic diseases.No history of previous use of special drugs,within one week before the sleep monitoring,upper respiratory tract infection or fever history was absent,quit smoking and alcoholl week,all patients were informed consent.2.Draw morning fasting blood 3ml from all the subjects,place the blood samples in abacterial blood vessels without DNA and RNA enzymes immediately after in vitro,let them stand naturally,then collect serum after centrifugeing 5 min,800×g,-80? saved for later use.3.Serum RNA extraction:take the serum samples,according to the methods described by the extracting kit instructions of miRcute miRNAs extraction.Using ultraviolet spectrophotometry to measure RNA concentration.4.MiRNA reverse transcription and synthesise to cDNA,Q-PCR amplification target gene to measure the miRNA levels.5.Collect the results,make the data analysis and statistical processing.Results:1.The expression of serum miR-181a in the mild OSAHS patients(10.5±0.5)compared with normal group(10.2±0.1),there is no statistically significant difference(P>0.05),but in the medium and severe OSAHS patients,the serum miR-181a has a higher expression level obviously(12.8±1.4),the difference was statistically significant(P<0.05).As the data are compared between groups,the expression level of serum in the medium and severe OSAHS patients are higher than the control group(P<0.05).It indicates that the serum miR-181a in OSAHS patients express in a higher level than the normal group.In addition,the serum miR-181a expression level relatively rise in severe OSAHS patients than the mild OSAHS group.(F=16.622,P<0.001).2.The expression of serum miR-181a in the OSAHS patients have a positive correlation with AHI and TS90%(r value are respectively 0.752 and 0.778,P<0.01).3.The expression of serum miR-181a in the OSAHS patients have a negative correlation with LSaO2(r value is-0.388,P<0.01).Conclusions:1.As the extent of OSAHS is aggravating,miR-181a with a higher expression level;With the increase of AHI and TS90%,miR-181a has a higher expression level;With the decreasing of LSa02 level,the expression level of miR-181a raise.2.MiR-181a is closely related to the PSG monitoring various indicators of OSAHS patients,their combined application can further improve the efficiency to the diagnosis and treatment of OSAHS,and help to assess the therapeutic effect of CPAP.
