Objective(s):With the gradual increase of the elderly population in our country,Alzheimer' s disease,as a kind of central nervous system degenerative disease,has became a hot topic in the world.Alzheimer's disease is a common type of senile dementia,with progressive memory loss,cognitive decline,mental and behavioral abnormality were the main clinical manifestations of the daily life of the elderly has brought serious damage,so the early identification,early diagnosis and intervention is particularly important for patients with dementia.In recent years there have been some reports that progranulin(Progranulin,PGRN)was associated with frontotemporal dementia,Alzheimer's disease,breast cancer,multiple sclerosis,systemic lupus erythematosus and other diseases,However,there is little study on the effect of the protein on Alzheimer's disease.According to the previous studies,some of the PGRN mechanism and the pathogenesis of Alzheimer's disease and vascular dementia,and PGRN can cross the blood-brain barrier,expressed in peripheral blood.Some research found that PGRN may participate in the pathogenesis of VD and AD,so we measured the serum in patients with several types of dementia PGRN protein concentration,and measured the MMSE and other related to cognitive scales to explore the correlation between the two.Methods:We selected 11 cases of Alzheimer's disease patients,20 cases of vascular dementia patients,24 cases of mixed dementia patients and 55 cases were matched by age and sex,using ELISA to determine the concentration of serum PGRN protein level by MMSE(Mini-mental state examination),ADAS-cog(Alzheimer's disease assessment scale cognitive section),ability of daily life(Activity of daily living,ADL)scale were used to assess the cognitive and memory of patients with dementia,using SPSS22.0 software package for general statistical analysis of statistical date and all inventory date using analysis of variance comparing four groups of rating scale and four groups of PGRN serum protein concentration difference,after inspection by using LSD-t method compared two further.The four groups of serum PGRN protein concentrations and ADAS scores,ADL scores,the detailed rules for the MMSE score,to conform to the normal distribution of date using Pearson correlation analysis,the date which does not meet the normal distribution of date Spearson method is analyzed.P<0.05 was considered statistically significant.Results:1.MMSE scale,ADL scale and ADAS between group and control group,the ADAS-cog rating scale,concentration of serum PGRN pairwise comparison,the team scale total score with the control(P<0.05),two scale scores between the two is found after the team each group:P were greater than 0.05,the team with the control group scale total score:P values are less than 0.05.AD group serum PGRN protein concentration(13.69±1.99)ng/ml,VD serum PGRN protein concentration is(19.39±1.34)ng/ml,MD serum PGRN protein concentration is(18.38±1.05)ng/ml,the control serum PGRN protein concentration is(20.39±1.69)ng/ml.2.The difference was statistically significant compared with the control serum PGRN protein concentration(F=59.36,P<0.01).The difference was statistically significant,comparing two further,[(MD group)vs(AD group)](P=0.000),[(VD group)vs(MD group)](P<0.05),[(VD group)vs(MD group)](P<0.05),[(control group)vs(VD group)](P<0.05).3.Team serum PGRN protein and MMSE,ADL,ADAS-cog scale score and total score of groups,P were greater than 0.05,further explore diverse research serum PGRN protein concentration and scale groupings,and the correlation between the total score,all P>0.05,but the AD group of MMSE scale of immediate recall a(P<0.05,r=0.037),the difference was statistically significant.Conclusion(s):1.There was no significant correlation between the scale scores of the groups,and the group's cognitive scale scores and serum PGRN concentrations were significantly lower than those in the control group.2.Serum concentration of PGRN:AD group<MD group<VD group<control group,serum PGRN protein may be involved in AD,VD,MD.3.The research team's serum PGRN protein was unrelated to MMSE,ADL,ADAS-cog scale ratings,but may be associated with an immediste recall of an AD patient.
