Lepidium latifolium L. is the Cruciferae perennial herbs of the genus Lepidium L.,and there are the corresponding records of Medicine in "Shen Nong’s herbal classic" which is the earliest traditional Chinese medicine monograph.Based on the study of pharmacodynamic active parts of Promoting gastrointestinal motility for Lepidium latifolium L.,in this study, the extracts are separated with different polar components,and the pharmacodynamic screening has been operated in the promotion of gastrointestinal motility,antimicrobial and antioxidant activities and other aspects,which aim to clear its effective components. It will provide the basis for making full use of this plant resources.In this study,by taking ethanol, butyl alcohol, ethyl acetate and petroleum ether as solvent,the extraction of Lepidium latifolium L. has been done.The ethyl acetate extract was eluted by taking chloroform methanol mixture system as the elution system.According to different polarity,it was separated to each component as the tested drugs.Then observed the effect of different components for gastrointestinal myoelectricity,amplitude and frequency of tension of rats in normal groups and model groups.At the same time,different components have been taken the antibacterial activity test to 13 strains including staphylococcus aureus,micrococcus luteus,etc,and have been taken the antioxidant activity test of scavenging DPPH free radicals.The results of the study are as follows:(1)The composition of chloroform methanol system(10:1 and 5:1) in ethyl acetate of Lepidium latifolium L. has significantly enhanced electrical activity(mv)of gastric and intestinal smooth muscle in vivo and the tension(g) of the stomach and intestines in vitro, which effect intensity is equivalent to the positive control group(Si Shah Billy).Among them, the effect of chloroform methanol(10:1)is better than that of the chloroform methanol(5:1) component.(2)The chloroform methanol(30:1, 20:1) components from the ethyl acetate extract of Lepidium latifolium L.,which has highly sensitive(Φ>15 mm),has goodantibacterial effect to Salmonella paratyphi,and its MIC for Salmonella paratyphi is respectively 1.25mg/ml and 0.625 mg/ml. The chloroform methanol(20:1)component from the butanol extract and ethyl acetate extract,which MIC for Escherichia coli is 2.5 mg/ml,has good antibacterial effect to Escherichia coli.And the butanol extract is moderately sensitive(Φ>10 mm).(3)Different extracts and components of Lepidium latifolium L. all have the ability to scavenge DPPH free radical.Especially,chloroform methanol(10:1)fractions of ethyl acetate extract,which value of SC50 is 61.27 μg /m L,has the strongest scavenging capacity.(4)Through the analysis and comparison of high performance liquid chromatography(HPLC),it shows that benzene C compounds and flavonoid compounds are the key ingredients from chloroform methanol(10:1) components in ethyl acetate extract of Lepidium latifolium L.,and the proportion of Benzene C compounds among them has reached 42.657%.The above results have indicated that the effective components of promoting gastrointestinal motility which are from ethyl acetate extract of Lepidium latifolium L.,mostly focus on the chloroform methanol(10:1, 5:1) fractions which effect is equivalent to cisapride.It indicates that it is the active component of promoting gastrointestinal motility for Lepidium latifolium L.;The active ingredients of anti Salmonella paratyphi B mostly focus on the chloroform methanol(20:1) component from ethyl acetate extract,and the active ingredients of anti escherichia coli mostly center on n-butyl alcohol extract.The chloroform methanol(10:1) fractions of ethyl acetate extract have good effect to scavenge DPPH free radical.
