The Study Of Pentraxin 3(PTX3)Gene Polymorphisms And Susceptibility Of Aspergillosis In Patients With Chronic Obstructive Pulmonary Disease

Chapter I The relationship between PTX3 polymorphism and susceptibility to Aspergillus in chronic obstructive pulmonary disease(COPD)ObjectionTo investigate whether PTX3 polymorphism was associated with Aspergillus infection among chronic obstructive pulmonary disease populationMethodsUsing case-control study.All participants enrolled were Chinese Han population and recruited in the Nanjing General Hospital of Nanjing Military Region between January 2015 and November 2016.The study comprised 173 unrelated subjects-36 pulmonary aspergillosis cases and 136 controls.2ml peripheral blood was collected from 173 patients.DNA samples were extracted.The PTX3 rs2305619,rs3816527,rs1840680 and rs3845978 polymorphisms were analyzed by direct sequencing.ResultsThis study comprised 173 subjects,36 patients with aspergillosis and 137 controls.There was no significant difference in gender,age,smoking history,history of hormone use,and severity of COPD between cases and controls.Among the four SNPs of PTX3 gene,Three SNPs(rs2305619,rs3816527,rs1840680)of the controls met Hardy-Weinberg equilibrium(HWE).The frequency of A allele in Aspergillus group rs1840680 was higher than that in COPD group.The difference was statistically significant(p=0.008).In the recessive model,the frequency of AA genotype was higher in the Aspergillus group than in the COPD group(p=0.00003).rs2305619,rs3816527 allele frequency and genotype frequency in the two groups were no significant difference between the two groups.ConclusionThe PTX3 rs1840680 polymorphism was associated with the occurrence of Aspergillus infection among COPD patients.rs2305619 and rs3816527 gene polymorphism showed no association with a risk of aspergillus infection.Compared with AG,GG genotype,AA genotype were the risk factors of Aspergillus infection.The haplotypes of rs2305619 and rs3816527 in PTX3 gene had no significant correlation with the occurrence of Aspergillus infection among COPD patients.Chapter ? Correlation between plasma PTX3 levels and incidence of aspergillosis in patients with chronic obstructive pulmonary disease of different PTX3 genotypesObjectionRelationship between plasma PTX3 levels and the occurrence of Aspergillus infection in different PTX3 genotypesMethods173 patients were collected using the case-control study.The patients were divided into COPD stable group,COPD acute exacerbation group and Aspergillusis group.Peripheral blood was collected from 173 patients.Separation of plasma and blood cells.Plasma levels of PTX3 were measured by enzyme-linked immunosorbent assay(ELISA).ResultsThe PTX3 values was not obviously different between COPD stages in COPD stable group.Compared with stable COPD and COPD acute exacerbation patients.plasma PTX3 levels were significantly higher in patients with aspergillosis.There was no significant difference in plasma PTX3 level between COPD stable group and COPD acute exacerbation group.The patients were classified according to rs1840680 polymorphism.There was a significant difference in plasma PTX3 level between AA genotype and AG/GG genotype in Aspergillusis group(p=0.039).ConclusionPlasma PTX3 levels were not associated with COPD severity in COPD stable group.Plasma PTX3 levels in patients with aspergillosis infection were significantly higher than in patients with COPD.This result suggests that plasma PTX3 has the potential to be a serum marker for chronic obstructive pulmonary disease with pulmonary aspergillosis.In patients with aspergillosis,the plasma PTX3 level of rs1840680 AA genotype was lower than AG/GG genotype.It was further confirmed that the rs1840680 A A genotype was a risk factor for Aspergillus infection.
