Expression Of TLR2?TLR3?TLR4 On Hepatocytes In Patients With Chronic Hepatitis B And Its Clinical Significance

Objective: We aim to investigate the association between the altered of toll-like receptor 2,3,4 in the iver tissue of chronic hepatitis B,finally got its clinical significance.Methods : We use a second quick puncture for liver biopsy method.10 normal liver tissues served as controls.The liver tissue of 156 CHB which were for TLR2,TLR3,TLR4 immunohistochemical.Then,we obtained the correlations of TLR2,TLR3 and TLR4 with their liver inflammatory activity and liver fibrosis degree.We use SPSS18.0 version statistical software to process data.Results:There are negative expression in control group.The expression of TLR2 in liver tissue of patients with CHB is weak,which is mainly expressed in the cytoplasm of liver cells at the cellular level,and the positive expression of liver nuclei is seen in one case.There was no correlation between TLR2 expression intensity and hepatic inflammatory activity scale(RS)(G:-0.086,P>0.05),and the expression intensity of TLR2 was not correlated with liver fibrosis grade(S):(RS value:-0112,P>0.05).TLR3 in liver tissues of patients with CHB high expression was mainly expressed in the cytoplasm of cells at the cellular level,a small part is generally expressed in the nucleus,cytoplasm or nucleus of simple expression,cell membrane staining.The expression of TLR3 in liver tissues and the inflammation grade(G)had a significant positive correlation(rs:0.528,P<0.01),and there is a significant linear relationship(?2:16.679,P<0.01).The expression of TLR3 grading of liver fibrosis and the degree of strength(S)had a significant positive correlation(rs:0.510,P<0.01),and there is a significant linear relationship between(?2:16.654,P<0.01).The expression of TLR4 is obvious,mainly localized in the cytoplasm of liver cells,some of which are expressed on the liver cell membrane,but not expressed in the nucleus.The expression of TLR4 in liver tissues and the inflammation grade(G)had a significant positive correlation(rs:0.414,P<0.01),and there is a significant linear relationship(?2:11.593,P<0.01),the expression of TLR4 grading of liver fibrosis and the degree of strength(S)had a significant positive correlation(rs:0.354,P<0.01),and a significant linear relationship(?2:10.124,P<0.05)Conclusions:(1)Liver tissue of patients with CHB on the expression of TLR2 is weak,there are no correlation with degree of liver tissue inflammation and fibrosis.(2)There are an increasing in the expression of TLR3 in liver tissue of patients with CHB,and the degree of liver inflammation and fibrosis was related to TLR3 activity,it may be involved in the pathogenesis of CHB,especially in immune injury.(3)There are an increasing in the expression of TLR4 in liver tissue of patients with CHB,and the degree of liver inflammation and fibrosis was related to TLR4 activity,it may be involved in the pathogenesis of CHB is involved in the formation of liver fibrosis,possibly associated with immune injury CHB.
