Effect Of Quercetin By Detect The Levels Of IL-1? And TNF-? In The Knee Joint Fluid Of Rosteoarthritis Rabbits

Objective:To observe the effect of quercetin by detect the levels of IL-1?and TNF-? in the knee joint fluid of rosteoarthritis rabbits.Methods :Experimental OA models of rabbit knee joint were established according to hulth method.Twenty New Zealand white rabbits were randomly divided into 4groups,Each group had 5 rabbits.(1)quercetin group(2)celecoxib group(3)saline group(4)model group.The four groups were received relevant medications once a day for 4 weeks after successful model establishing.The quercetin group was administrated with quercetin with 10.31 ml per kg by gastric perfusion,;equivalent celecoxib was intragastrically administrated in the celecoxib group;equivalent saline was tintragastrically administrated in the saline group.While the model group would not received any treatment.After four-weeks treatment,the rabbits were killed by air embolism,Then the knee joint fluid was extracted,the IL-1? and TNF-? levels in the knee joint fluid were measured by ELISA method.and the articular cartilage of the femoral medial condyle was sent to the common pathological examination,The general scorewere recorded to estimate degree of knee joint damage.Results:(1)Cartilage surface exfoliation can see in the rabbits condyle in model group,the depth is to the bone,osteophyte formation in rabbits articular.bone tissue and cartilage cells are less through HE.(2)The quercetin and celecoxib group had a significantly improvements than model group and saline group in general knee joint score(P<0.05),The quercetin and celecoxib group showed no obviously changes(P>0.05).(3)The quercetin and celecoxib group had a significantly improvements than model group and saline group in the levels of IL-1? and TNF-?(P<0.05).Conclusions : Quercetin and celecoxib could significantly inhibit articular cartilage destruction and decreased the levels of IL-1? and TNF-? in synovial fluid of OA model.
