Objective:Human periodontal ligament cells(hPDLCs)Cultured in vitro,would secret Tumor necrosis factor-?(TNF-?)and Interleukin-6(IL-6)at different points,after stimulation of high glucose,Porphyromonas gingivalis(P.g),bacteria Lipopolysaccharide(LPS)by cenzyme-linked immunosorbent assay(ELISA).We examined the changes and significance of secretion.This study is to explore the function and mechanism of PDLCs stimulation by LPS,where inflammatory cytokines would be elevated.Providing theoretical basis for immune pathological mechanism of diabetes mellitus and periodontitis in the discussion.It is this research that we probe new ideas and methods for diabetic periodontitis with inflammation controlled and tissue repaired.Methods:1?Healthy patients(aged 12 to 18)chosen,were extracted premolars with noncaries,non-periodontitis in the need of orthodontic treatment.After informed consent,periodontal membrane was obtained immediately in the one-third of root after extraction.4-6 generations of cells would be applied in the experiment,after Primitive cultivation of hPDLCs by tissue blocked method.2?The study is aimed to detect secretion of IL-6 and TNF-? under high glucose by ELISA,after stimulation of Pg LPS(10?g/ml)6?12 hours later respectively,in the comparison of blank controlled group(glycopenia)and Pg LPS group(glycopenia)Results:1?Original generation cultivated a week later,some cells migrated from edge of tissue microscopically,they are long fusiform or star,with 2 to 4 cytoplasm,surrounding tissue block stellately,part of them as woven mesh.2 weeks later they migrated from tissue to full of the whole bottle.After it was full of bottle,we cultivated them in generations,4-6 generations of periodontal ligament fibroblasts applied in the experiments.2?In the state of high glucose ?LPS and joint stimulation 6 hours later,TNF-? secretion of hPDLCs significantly higher than the blank control group(P < 0.05).As time goes on,the effect is more evident,and it is elevated as time flies.Especially in the high glucose and LPS joint stimulation 12 hours later,periodontal ligament fibroblasts produce TNF-? were significantly higher than that of the rest of the group.3?After high glucose stimulation 6 hours later,compared with blank control group the hPDLCs secrete IL-6 had no significant changes,with no statistical difference(P > 0.05).However,LPS and LPS + high glucose stimulation of the two groups compared with the blank control group(P < 0.05),significantly increased the secretion of IL–6.AS time goes on,the experimental results with statistical difference,as time go on.Conclusion:1?the state of high glucose can facilitate hPDLCs to secret TNF-??IL-6.2?the state of P.g LPS can facilitate hPDLCs to secret TNF-??IL-6.3 ? the state of high glucose can significantly facilitate the hPDLCs under inflammation to secret TNF-??IL–6,to a certain extent,reflecting the hyperglycemia of diabetic can increase the immune response of periodontitis.
