Objective To study the effects of Astragaloside(AS)on periodontitis in diabetic rats and its correlation with inflammatory factors,providing new drug targets for the clinical treatment of diabetic periodontitis.Methods Streptozotocin(STZ)was used to induce the diabetic rat model of diabetes mellitus with high glucose and high fat diet.On this basis,the maxillary molars were inoculated with porphyromonas gingivalis ligation causes periodontitis,resulting in a DP rat model.The N group,the DP group and the AS treatment group were tested.Comparison of blood glucose,gingival inflammation,periodontal pocket depth,alveolar bone loss and other related indicators;observation of blood-biochemical indicators: AGEs,C peptide,Hb Alc changes;the levels of IL-1,IL-6 and TNF-? in alveolar bone of four groups were detected by immuno histochemistry.Comprehensive analysis of the effects of AS in diabetic periodontitis and the correlation between inflammatory factors.Results1.Blood glucose,urine sugar and other changes: successful model of DM rats fasting blood glucose were higher than 16.7mmol/L,significantly higher than the N group,the difference was statistically significant(p<0.05).The levels of blood glucose and urine glucose were negative or weakly positive in the high and low dose group treated with AS,and thedifference was statistically significant(p <0.05).2.Periodontal examination of AS group showed a decrease in the performance of gingival inflammation,periodontal pocket shallow,smaller attachment loss level,DP group of severe gingival inflammation in rats,periodontal bags to explore the deeper,attached to the loss of a large level,N group was normal,the two pairs were statistically significant(p<0.05).3.In the ipsilateral periodontal tissues,paraffin sections were observed.HE staining showed a large number of inflammatory cells in the periosteal bag of DP group.The maxillary bone samples showed horizontal absorption of alveolar bone.AS group showed that the degree of infiltration of epithelial inflammatory cells in the periodontal pocket was lower than that in the DP group.N group without the above changes.4.Biochemical analyzer was used to detect the levels of AGEs and Hb A1 C in AS treatment group,the content of serum C peptide increased,DP group AGEs and Hb A1 C levels were higher,serum C peptide content was low,N group were in normal level,the difference between them were statistically significant(p <0.05).5.The levels of IL-1,IL-6 and TNF-? in the serum of AS group were decreased by enzyme-linked immuno-sorbent assay(ELISA),the levels of IL-1,IL-6 and TNF-? in the serum of DP group were higher than those in the control group,and the difference was statistically significant(p <0.05).Conclusions1.Astragaloside has hypoglycemic function.2.AS could reduce the level of AGEs and Hb A1 C,increase the content of C peptide in DP rats,promote periodontal tissue repair.3.Astragaloside might reduce the inflammatory mediators,control inflammation of the alveolar bone damage,could relieve the inflammation and destruction of diabetic periodontitis.
