Objective:The mRNA and protein expression level of IL-34、IL-6、M-CSFR in gingival tissues of chronic periodontitis patients and healthy people were detected.to study the correlation between IL-34、IL-6、M-CSFR cell levels and clinical examination indexes of periodontal disease in each group..To evaluate the development and prognosis of chronic periodontitis and provide reference materials for the treatment of chronic periodontitis,the possible mechanism of IL-34、IL-6、M-CSFR in the occurrence and development of chronic periodontitis was preliminarily discussed.Methods:30 Patients who were diagnosed with chronic periodontitis in the department of stomatology,zhejiang hospital of traditional Chinese medicine from March 2018 to August 2018 were collected,including 15 mild to moderate patients,15 severe patients and 15 healthy patients as the control group.Patients’s name,age,gender,PD,AL,GI SBI were recorded in detail.Real-time PCR(rt-pcr),Elisa,Western blot were used to detected the mRNA and protein expression level of IL-34,IL-6 and M-CSFR in ginggival tissue from those samples.SPSS 17.0 statistical method was used to analyze the experimental results,and p<0.05 was considered statistically significant.Results:1.The result of PCR showed that the expression of IL-34.IL-6.M-CSFR were significantly higher in chronic periodontitis than in normal tissues.But there was no significance difference in the expression of M-CSFR between slight or moderate peiodontitis gruop and severe peiodontitis gruop(p>0.05).2.The result of Western blot showed that the expressions of IL-34、IL-6、M-CSFR in chronic periodontitis were significantly higher than controls(p<0.05),and severe peiodontitis gruop was higher than slight or moderate peiodontitis group(p<0.05).3.The result of Elisa showed that the expression levels of IL-34、IL-6、M-CSFR increased with the increase of periodontal inflammationr.The expression of IL-34 was statistically significant only in the case of severe periodontitis(p<0.05),compared with the control group,whereas there were no statistically significant between severe peiodontitis gruop and slight or moderate peiodontitis group(p>0.05).The expression of IL-6 and M-CSFR were statistically significant when compared between mild,moderate and severe groups.4.Clinical examination indexes of periodontitis,including periodontal pocket depth,loss of attachment,gingival index and gingival groove bleeding index,were positively correlated with il-6,il-34 and m-csfr in mRNA level.Conclusion:The mRNA and protein levels of IL-34、IL-6、M-CSFR in gingival tissue of chronic periodontitis were significantly higher than controls(p<0.05).It is also significantly correlated with the clinical examination indexes of periodontitis:periodontal pocket depth,loss of attachment,gingival index and gingival groove bleeding index.It was found that the change of its content was related to the degree of inflammation in periodontal tissue.
