The Inhibition Effects And Mechanisms Of The Morusin On The MDA-MB-453,HCT116 And H1299 Cancer Cells

Abstract/Summary:
In this experiment,we used morusin as the experimental material.We found that morusin had the growth inhibition on most cancer cells through the drug screening of 20 kinds of cancer cells.Therefore,we investigated the effects of morusin on the proliferation,invasion and apoptosis of breast cancer cells MDA-MB-453,human lung cancer cells H1299 and colon cancer cells HCT116,and explored its mechanism initially.The results laid the foundation for the sustainable utilization of silkworm waste resources,the sustainable development of silkworm industry and the anti-tumor of traditional Chinese medicine.And it’s also provide the necessary theoretical and experimental basis for the research and clinical application of bioactive substances-morusin.The main contents of this paper are as follows:(1)In this paper,20 kinds of cancer cells were screened by morusin.The control group(0.5%DMSO)and different concentration of morusin-treated groups(1.25 ^gg/ml,2.5μg/ml,5μg/ml,10 μg/ml,20 μg/ml)were used in this experiment.The results showed that morusin can inhibit the growth of 20 kinds of cancer cells.The tumor inhibitory rate of high concentration treatment group(20 μg/ml)could reach more than 84%and showed a dose effect.IC50 results showed that 20 kinds of cancer cells’ IC50 average was 6.21μg/ml,of which 80%of the cells IC50 was less than or equal to 6.45μg/ml.It cloud be concluded that morusin had a very good inhibitory effect on most cancer cells.(2)In this paper,we used breast cancer cell MDA-MB-453,human lung cancer cell H1299 and colon cancer cell HCT116 as experimental materials,and treated with different concentrations of morusin(1.25 μg/ml,2.5 μg/ml,5 μg/ml,10 μg/ml,20μg/ml)for 24h,48h and 72h,respectively.The results showed that morusin had inhibitory effect on the growth of the above three kinds of cells,and showed time-concentration-dependent.The inhibitory rate of above three kinds of cells treated with 20 μg/ml morusin for 48h was 94.02%,97.9%,91.9%,respectively.(3)We treated the breast cancer cell MDA-MB-453,human lung cancer cell H1299 and colon cancer cell HCT116 with different concentrations of morusin(2.5μg/ml,5μg/ml,10μg/ml).Western blotting showed that morusin could up-regulate the expression of E-cadherin,down-regulate the expression of N-cadherin and Vimentin,and showed a dose effect.The results showed that morusin can reverse EMT,inhibit tumor migration,invasion and infiltration.(4)We treated the breast cancer cell MDA-MB-453 and colon cancer cell HCT116 with different concentrations of morusin(5μg/ml,10μg/ml,20μg/ml)for 48h.The results showed that the cells were apoptosis,and showed dose-dependent.The apoptotic rate of MDA-MB-453 cells treated with high concentration(20μg/ml)of morusin was 77.73%(P<0.01).Western blotting implied that morusin could up-regulate the expression of Bax and cleaved caspase-3,down-regulate the expression of Bcl-2 and caspase-3,indicating that morusin could induce cell apoptosis.(5)We treated the breast cancer cell MDA-MB-453 and colon cancer cell HCT116 with different concentrations of morusin(2.5μg/ml,5μg/ml,10μg/ml).The results showed that MDA-MB-453 cells were arrested in G2 phase and HCT116 cells were blocked in S phase when the concentration of morusin was 10μg/ml.Consequently,morusin could disturb the cell cycle.
