Objective: To explore the effect of a defined formula of Chinese medicinal herbs-YankeningⅣ(also known as the pelvic inflammation relieving formulaⅣ) on the model rats with chronic pelvic inflammatory disease(CPID) by observing the roles that it may play in the processes of immunoregulation and the control of cell apoposis.Methods: The models of rats with CPID(Wistar female rats) were successfully made by infecting three various bacteria into the rats. Sixty-seven female Wistar rats were randomly classified into the following different groups: the normal group, the simulative model group, the model group, the high dose of YankeningⅣgroup, the medium dose of YankeningⅣgroup, the low dose of YankeNingⅣgroup and the control group with another kind of Chinese medicinal formula.The light microscope and electron microscope were used in the morphologic research and the method of immuno-histochemistry was used to detect the expression of INF-γand Caspase-3 in the uterine tissues of the model rats with CPID. The method of enzyme linked immunosorbent assay(ELISA) was used to detect the content of IL-2 and IL-6 in the serum of the model rats. The method of reverse transcription follow by PCR(RT-PCR) was used to detect the expression of INF-γmRNA, MMP-2mRNA, Fas/FasLmRNA and p53mRNA in the uterine tissues of the model rats with CPID.Results: The contents of IL-2 and IL-6 in the serum of the model rats were significantly higher than those of the normal rats(P<0.05), so were the expressions of INF-γin the uterine tissues of the model rats with CPID(P<0.05). The expressions of Caspase-3, Fas/FasLmRNA, p53mRNA and MMP-2mRNA in the uterine tissues of the model rats were significantly weaker than those of the normal rats(P<0.05). YankeningⅣhas significantly decreased the contents of IL-2 and IL-6 in the serum of the model rats and the expressions of INF-γin the uterine tissues of the model rats with CPID(P<0.05). It has also significantly increased the expressions of Caspase-3, MMP-2mRNA, Fas/FasLmRNA and p53mRNA in the uterine tissues of the model rats(P<0.05). The high dose of YankeningⅣgroup functioned significantly better than the control group with another kind of Chinese medicine(P<0.05).Conclusions: It was through decreasing the contents of IL-2 and IL-6 in the serum of the model rats and the expressions of INF-γin the uterine tissues of the model rats with CPID that YankeningⅣplay the roles of restore the immune disequilibrium,thus relieving the pelvic inflammation. It was through increasing the expressions of MMP-2mRNA in the uterine tissues of the model rats that YankeningⅣdegrades extracellular matrix and inhabits the fibroplasia and then facilitate to remove CPID. It was through increasing the expressions of Caspase-3, Fas/FasLmRNA and p53mRNA in the uterine tissues of the model rats that YankeningⅣpaly the roles of inducing the programmed inflammation cell death, thus relieving and removing CPID. YankeningⅣis an kind of effective defined formulas of Chinese medicinal herbs in treating CPID. |
