Objective: micro RNAs(miRNAs)is a kind of endogenous,single stranded,conserved,non coding RNA,which regulate cell proliferation,differentiation and apoptosis through negative regulatory gene expression in post-transcriptional levels.Because of its concentration in these processes,mi RNAs play an important role in the process of human tumor,and are closely related to tumor occurrence,development,diagnosis and prognosis.The relevant literature and experimental data suggest that miRNAs have certain expression in papillary thyroid carcinoma(PTC),which is valuable and instructive in the diagnosis and treatment of PTC.But its mechanism is still unclear,and the relationship between clinical and pathological features to be further studied and analyzed.The purpose of this study is to validate the expression of miRNAs in PTC,to screen for miRNAs with different expression and to analyze their clinical correlations to distinguish PTC and benign tumors,to develop,improve and enhance the diagnosis and treatment strategies of PTC.Methods : Collected the thyroid papillary carcinoma,benign tumor tissue,adjacent to the tumor distal non-tumor thyroid tissue(distance tumor Distal age non-normal thyroid tissue)of 65 patients,which in the department of vascular and thyroid surgery inSouthwest Medical University from June 2015 to December 2016.Among them,30 cases of thyroid papillary carcinoma,11 males and 19females;aged 13-71 years,mean age 46.08 ±15.68 years;15 cases of tumor diameter more than 2cm,15 cases of tumor diameter less than 2cm(the diameter of tumor pathology report to date);13 cases of multifocal carcinoma,17 cases of single cancer;there were 19 cases of lymph node metastasis,11 cases without lymph node metastasis;ATA PTC recurrence risk classification: 18 cases of low risk group,12 cases of high risk group;TNM classification: 19 cases of stage I,11 cases of II-IV patients;25cases of the traditional type of PTC,5 cases of follicular papillary carcinoma patients;15 cases of thyroid globulin Tg ? 77ng/ml,Tg >77ng/ml;5 cases of papillary thyroid microcarcinoma.There were 35 cases of benign thyroid tumor,24 cases of nodular goiter,30 cases of thyroid adenoma,5 males and 30 female,with an average age of 46.03±13.40 years old.The specimens were taken in the operation of non electric knife burning fresh tissue,obtained immediately stored in liquid nitrogen tanks,and quickly transferred to-80? refrigerator to keep alive.All specimens of patients underwent surgical resection were determined by routine histopathological examination.By referring to the TCGA data and combining with the relevant literature,we selected a total of 8miRNAs,which expressed in PTC but not further validated.It included miR-532,miR-455,miR-551,mi R-1287,miR-1975,mi R-1976,miR-2355,miR-1266,detecting the expression by RT-qPCR in PTC,benign tumor and adjacent to the tumor distal non-tumor thyroid tissue to analysis the correlation between the clinical features of thyroid papillary carcinoma.Results: The results showed that the expression of miR-1287,miR-1975,miR-1976,miR-2355 and miR-1266 in thyroid papillary carcinoma tissues had some changes,but there was no significant statistical significance.Compared with the normal tissues,the expression of miR-532 in benign and malignant tumors was significantly decreased,but there was no significant difference between the benign and malignant tumors;compared with the normal tissues,the expression of miR-455 in benign and malignant tumors was significantly increased,but there was no significant difference between the benign and malignant tumors;compared with the normal tissues and benign tumors,the expression of miR-551 in malignant tumors was significantly increased.Correlation analysis of clinical and pathological features revealed that the expression of miR-455-3p was correlated with TNM stage,tumor size and age.The expression of miR-532-5p was correlated with age,lymph node metastasis and TNM stage.The ROC analysis showed that miR-455-3p,miR-551-3p,mi R-532-5p has a certain value in diagnosis of thyroid tumors,and miR-455-3p is the most suitable for distinguishing tumor tissue from normal tissues and its sensitivity was 75.0% and the specificity was 86.4%.Conclusion: The expression levels of miR-455-3p,miR-551-3p and miR-532-5p in PTC were significantly correlated with clinical and pathological features,such as age,lymph node metastasis and TNM stages,and they may play an important role in the diagnosis of thyroid papillary carcinoma,preoperative evaluation and improvement of treatment strategy.
