The Mechanism Study On Kidney-yang Deficiency And Constipation Cured By Warming Yang For Relax Bowels

Objective:To apply activated carbon combined ice water and 5%of acetic acid to make animal model of rats' kidney-yang deficiency and constipation and discuss the relation between kidney-yang deficiency and constipation cured by warming Yang for relax bowels and MTL,SP and SS,respectively.Methods:60 SD rats were lavaged for 4 weeks by activity carbon ice water combined with 5%of acetic acid and rat models with kidney-yang deficiency and constipation were made.High-dosage group(3.8g/kg),middle-dosage group(1.9g/kg)and low-dosage group(0.95g/kg)of Jichuan Decoction were prepared for the concentration of 25.2%,12.6%and 6.3%,respectively.Rats were provided with intragastric administration of 1.5ml/100g.The model groups gave normal saline with the same volume for once a day for 7 days.General physiological change of rat's first defecate time and discharge quantity within 6 hours,as well as myoelectricity and pathological section were observed.Moreover,Elisa and immunohistochemistry were used to detect MTL,SP and SS level in rats' serum and colon.Results:Experimental groups of colon pathological section had no difference.After modeling,general physiology of rats' characteristics include abdominal distension,hard fur,cowered body,light weight,and black and dried defecate.Compared with the control group,rats in model groups had longer executive time.The number of discharge particles was reduced.Gastrointestinal motility was weakened.MTL level in serum and colon was reduced.P has no constant substances,and SS content was rising.The above-mentioned signs and indicators in the treatment group were improved obviously.Moreover,high-dosage group and middle-dosage group of Jichuan Decoction had more obvious curative effects than low-dosage group of Jichuan Decoction.Conclusions:After activated carbon combined ice water and 5%of acetic acid model,colon had no organic cause lesions and belonged to functional constipation.Signs of rats conformed to kidney-yang deficiency.The created rat constipation model was the model of kidney-yang deficiency and constipation.The Jichuan Decoction cured kidney-yang deficiency and constipation had a close relation with the improvement of MTL level and reduction of SS level,but had no relation with P substance.Moreover,high-dosage group and middle-dosage group of Jichuan Decoction had more obvious curative effects than low-dosage group of Jichuan Decoction.
