Objective: To study and compare the evaluation of quality,four kinds of chemical constituents,qualitative and quantitative analysis of trace elements determination in Folium Eucommiae from different geographgic regions,evaluate Folium Eucommiae quality.Methods: According to the Pharmacopoeia of the people's Republic of China and literature character identification of Folium Eucommiae,It's powder for microscopic identification,for TLC identification of the Folium Eucommiae from 7 batches in the chemical composition of chlorogenic acid.Determine the content of total polysaccharides,total flavonoids,total polyphenols in the Folium Eucommiae from 7 batches by vis-spectrophotometry.Applied fourier transform infrared(FT-IR)Spectroscopy to study the Folium Eucommiae to establish the IR fingerprint.The use of high performance liquid chromatography(HPLC)techniques to analyze four chemical constituents in the Folium Eucommiae from 7 batches.Applied flame atomic absorption spectrometry(FAAS)method and Graphite furnace atomic absorption spectrometry method to determine nine trace elements in the Folium Eucommiae from 7 batches,soil samples and the irrigation water samples.Result: The optimized TLC condition is perform on silica gel H TLC plates with butyl acetate-formic acid-water(7:2.5:2.5,volume ratio)as mobile phase,With this method,the Folium Eucommiae from 7 batches were analysed and found compounds were the same.The content of Total polysaccharide,total flavonoids and total polyphenol in the Folium Eucommiae from 7 batches was different,among them,sample S7 had higher contents.According to the information of IR peak shape and peak position,the characteristic of spectum have not distinguished for the Folium Eucommiae from 7batches until the second derivative infrared spectra was used.The second derivative IR can enlarge the differences between samples.Five groups were classified according to cluster analysis for Folium Eucommiae.The content of chlorogenic acid,quercetin,kaempferol and rutin is different in the Folium Eucommiae from 7 batches,the content of chlorogenic acid is larger.The content of potassium and calcium was generally high in the Folium Eucommiae from 7 batches,soil samples,irrigation water samples,heavy metal elements are in line with national standards.Conclusion: This study explore and compare the content of total polysaccharide,total flavonoids and total polyphenol in the FoliumEucommiae of Xinjiang production and other 4 areas Folium Eucommiae.To the character identification,microscopic identification and TLC for the Folium Eucommiae from 7batches.Using of HPLC techniques to analyze four chemical constituents in Folium Eucommiae from 7 batches.Determination the content of trace elements in the Folium Eucommiae from 7 batches,soil samples and irrigation water samples.For the establishment of a scientific quality standard of Folium Eucommiae and provide scientific basis for the Folium Eucommiae of Xinjiang production of medicinal resources widely development and utilization.
