Objectives:Eucommiae cortex is the bark of Eucommia ulmoides oliv, It possesses the function of strengthening bone, tonifying liver, regulating blood-lipid, lowering blood pressure, anti-oxidation activity and antimicrobial activity. Eucommiae cortexs on the market include spotty products, Chinese Pharmacopoeia (2010) using HPLC method determination of pinoresinol diglucoside in eucommiae cortex as its quality control method, which is uncomprehensive. To provide theoretical foundation for the research of quality standards of eucommiae cortex, research on the content of this paper is mainly referred to the following three parts:establishing and improving a quantitative assay of Multi-components by Single-marker for the determination of geniposidic acid (GPA), chlorogenic acid (CHA), geniposide (GP) and pinoresinol diglucoside (PDG) in eucommiae cortex; establishing near-infrared spectroscopy quantitative analysis of main bioactive components in eucommiae cortex using partial least squares (PLS); establishing standard fingerprints for cortex eucommiae and providing an effective method for quality control.Methods:1. With CHA as reference substance, we calculated the relative correction factor of GPA, GP and PDG from the ratio of slope of calibration curve. using the area of each peak and the relative correction factors, the contents of GPA, GP, PDG in cortex eucommiae were determined;2. Near infrared diffusere flectance spectra were collected from eucommiae cortex powder and pretreated by chemometrics methods. A regression model between the content data from HPLC and the NIR data has been built up by PLS;3. We established standard HPLC fingerprints of eucommiae cortex growing in all parts of the country and Zhangjiajie respectively, the fingerprints of cortex eucommiae were comparatively analyzed using chemometrics methods.Results:1. A comparison of the results of external standard method and QAMS for the determination of GPA, GP and PDG indicates that the correlation coefficients for the two results are greater than0.99.2. The results showed that the model of the content of PDG reached to0.9615of the correlation coefficient between the prediction and the measured values, the RMSECV is in the range of0.0015; The model of the content of GPA reached to0.9583of the correlation coefficient between the prediction and the measured values, its RMSECV is in the range of0.0064.3. Result shows the similarity of all eucommiae cortex samples is in the range from0.76to0.98, and PCA results map respectively get Hubei, Yunan, Hebei, most of the Zhangjiajie sample together. Partial least squares method-discrimination analysis (PLS-LDA) methods combined with the obtained HPLC fingerprints were employed to build up the discriminant model between Zhangjiajie and other samples. Six compounds were selected as the potential markers in the cortex eucommiae from Zhangjiajie via Competition adaptive weighted samples (CARS).Conclusions:The improved QAMS of establishing slope correction is accurate and cheap, it can be used for analyzing synchronously the content of GPA, CHA, GP and GPA in cortex eucommiae. NIR spectroscopy analysismodel is very convenient and has good potential to quantitative detect active ingredients in eucommiae cortex. eucommiae cortex growing in zhangjiajie has its characteristic and the six iconic compounds can be as scientific basis for applications and Protection of Geographical Indication Products.
