Objective:Airway remodeling is closely related to repeated injury and repair of airway wall caused by airway inflammation,and “Qi deficiency,phlegm,blood stasis” are the important pathogenesis of COPD airway remodeling,among this qi deficiency runs through the whole process of the occurrence and development of COPD.This experiment selects a traditional Chinese medicine practitioner of Yunnan Province,Professor Li Qingsheng's prescription “Yupingfengsanflavored”,observing the changes of airway inflammatory microenvironment related factors,in order to discuss the effect of this prescription on improving COPD inflammatory microenvironment and inhibit airway remodeling process.Methods: 1.Grouping:SPF grade Wistar rats,weighing 180 ~ 220 g,male,divided into the normal control group,the model group,the positive drug group,the Yupingfengsanflavored low,medium and high dose group,the Yupingfengsan low,middle and high dose group,Yiqi couplet medicineslow,middle and high dose group.Simulated clinical disease development process set up: prevention of drug delivery group,drug delivery afterthe mode set upgroup,4 weeks after the model of drug delivery group,8 weeks after the model of drug delivery group,in addition to the prevention of drug delivery group didn't set up positive drug group;2.Copying model: COPD model rats were reconstructed by dripping LPS applied to trachea twice combined with passive smoking,and the modeling time is 8 weeks;3.Index observation: Observing the behavior changes of rats and pathological changes of lung tissues,and using ELISA to detect the changes of sIgA,SC,TNF-?,IL-8 expression levels,using 16 S rDNA to sequence the variable regions(V3,V4,V5),and the diversity of airway microflora in rats was analyzed by Illumina MiSeq sequencing platform.Results: 1.The HE staining results of pathological sections of lung tissue in rats:Traditional Chinese Medicine groups:The lung bronchial epithelium of rats had a decrease in proliferation,inflammatory cell infiltration around the wall and pulmonary interstitium,which was improved compared with model groups,and the effect of Yupingfengsanflavored of the 4 weeks after the model of drug delivery phase was better.2.The changes of sIgA,plgR / SC,TNF-?,IL-8 expressions of rats:(1)The prevention of drug delivery group:The model group :the expression levels of sIgA and IL-8 of decreased,SC and TNF-? expression levels increased;Traditional Chinese Medicine groups:In addition to the Yupingfengsan middle dose group,the expression levels of sIgA in the other Traditional Chinese Medicine groups were increased,and the increase of Yiqi couplet medicines middle dose group was the most obvious;The SC expression levels of Yupingfengsanflavored low middle and high dose group,Yupingfengsan low middle and high dose group,Yiqi couplet medicines high dose group decreased,Yiqi couplet medicines low and middle dose group increased,and the increase of Yiqi couplet medicines low dose group was the most obvious;The TNF-? expression levels ofYupingfengsanflavored middle dose group,Yiqi couplet medicines low,middle and high dose groupdecreased,Yupingfengsanflavored low and high dose group and Yupingfengsan low,middle and high dose groupincreased,and the decreaseof Yiqi couplet medicines middle dose group was the most obvious;The IL-8 expression levels of all Traditional Chinese Medicine groups were decreased,and the decreaseof Yupingfengsan low dose group was the most obvious.(2)The mode set up group:The sIgA,SC and TNF-?expression levels did not change significantly,the expression level of IL-8 of the model group decreased,all Traditional Chinese Medicine groups were decreased,and the decreaseof Yupingfengsan middle dose group was the most obvious.(3)4 weeks after the model of drug delivery group:The model group :the expression levels of sIgA and IL-8 of decreased,SC and TNF-? expression levels increased;Traditional Chinese Medicine groups:The sIgA expression levels of Yupingfengsanflavored low dose group,Yupingfengsan middle dose group,Yiqi couplet medicines low middle and high dose group increased,Yupingfengsanflavored middle and high dose group,and Yupingfengsan low and high dose group decreased,the increase of Yupingfengsan middle dose group was the most obvious;The SC expression levels of Yupingfengsanflavored low and middle dose group,Yupingfengsan low middle and high dose group,Yiqi couplet medicines low middle and high dose group decreased,Yupingfengsanflavored high dose group increased;In addition to the Yupingfengsanflavored high dose group and Yiqi couplet medicines middle dose group,the expression levels of TNF-?in the other Traditional Chinese Medicine groups were decreased,and the decrease of Yupingfengsanflavored middle dose group was the most obvious;The IL-8 expression levels of all Traditional Chinese Medicine groupsdecreased,and the decreaseof Yupingfengsan high dose group was the most obvious.(4)8 weeks after the model of drug delivery group:The model group :the expression levels of sIgA and IL-8 of decreased,SC and TNF-? expression levels increased;Traditional Chinese Medicine groups:The sIgA expression levels of all Traditional Chinese Medicine groupsincreased,the increase of Yiqi couplet medicines middle dose group was the most obvious;The SC expression levels of all Traditional Chinese Medicine groupsincreased,the increase of Yiqi couplet medicines middle dose group was the most obvious;The TNF-?expression levels of all Traditional Chinese Medicine groupsdecreased,the decrease of Yupingfengsan middle dose group was the most obvious;The IL-8 expression levels of all Traditional Chinese Medicine groups were decreased,and the decreaseof Yiqi couplet medicines middle dose group was the most obvious.3.Composition and variation of airway flora:(1)The prevention of drug delivery group:The contents of Proteobacteria,Firmicutes and Bacteroidetes in the model group decreased,and the contentsof all Traditional Chinese Medicine groups still decreased;The contents ofPseudomonas,Sphingobium,Rhizobium,Acinetobacter in the model group decreased,and the contentsof all Traditional Chinese Medicine groups still decreased;(3)4 weeks after the model of drug delivery group:The contents of Proteobacteria,Firmicutes and Bacteroidetes in the model group decreased,Traditional Chinese Medicine group: The contents of Proteobacteriadecreased,Firmicutes and Bacteroidetesincreased;The contents ofPseudomonas,Rhizobium,Acinetobacter in the model group increased,Sphingobium decreased,Traditional Chinese Medicine group: The contents ofPseudomonas,Rhizobium,Acinetobacter in the model group decreased,the content ofSphingobiumin Yupingfengsanflavored high dose groupincreased,Yupingfengsan high dose groupdecreased.It was suggested that Yupingfengsanflavored could regulate the contents of Proteobacteria,Firmicutes,Bacteroidetes,Pseudomonas,Rhizobium,Acinetobacterand Sphingobium.Conclusion:1.Yupingfengsan flavored can regulate the expression levels of sIgA,plgR / SC,TNF-?,IL-8,and slow down the development of airway inflammation;2.Yupingfengsan flavored can affect the airway flora structure and abundance,and reduce airway inflammation level;3.Yupingfengsan flavored can regulateairway inflammatory micro-environment balance,which may be one of the mechanisms of this prescription to delay the process of COPD airway remodeling.
