Experimental Study On The Effect Of Yupingfengtian Siwu Decoction And Yiqi Medicine On Intervention Of COPD Airway Remodeling

Objective: To observe the effects of Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine on related factors of airway remodeling in COPD rats, so as to investigate the part effect mechanism of Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine in the treatment of COPD.Methods: 1. The total of 350 Wistar male rats, and randomly assigned to four periods,each period were randomly divided into normal control group, model control group,positive drug control group(except for period 1), Yupingfeng Powder combined with Siwu Decoction high, medium, low dose group and Yiqi Couplet Medicine high,medium, low dose group, with 10 rats in each group; the period 1 smoked 28 days,period 2 smoked 56 days, periods 3 smoked 84 days, period 4 smoked 112 days, each period were administered for 28 days.2. The COPD rat model was reproduced by the method of smoke combined with instillation of lipopolysaccharide.3. The contents of SIg A, SC, TGF-β1, VEGF, MMP-9 and TIMP-1 in rat lung homogenate were detected by ELISA, and the pathological changes of lung tissue were observed by HE staining.Results: 1. Each period of each drug group can be different degrees of decrease of SIg A,SC, TGF-β1, VEGF, MMP-9 and TIMP-1 levels, elevated content of MMP-9 and MMP-9 / TIMP-1 ratio; which Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine low dose group and positive drug control group on the index of the most obvious(P<0.01), the difference is statistically significant.Conclusions: 1. Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine in treating COPD model rats, the peribronchiolar inflammatory cell infiltration, airway wall fibrous tissue hyperplasia, airway fibrosis and thickening of the wall, luminal stenosis and emphysema is reduced.2. One of the mechanisms of Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine in treating airway remodeling in COPD may is by reducing the level of TGF-β1 and VEGF and regulation of MMP-9 / TIMP-1 ratio imbalance, inhibit the growth of fibroblasts and endothelial cells, to further reduce the proliferation of bronchial smooth muscle cells, reduce vascular wall thickening,stenosis, degradation of bronchi, alveolar wall matrix deposition, so as to alleviate the airway remodeling in COPD.3. Yupingfeng Powder combined with Siwu Decoction and Yiqi Couplet Medicine has some effect on the development of blocking the airway remodeling in COPD. The low dose namely clinical equivalent dose was most obvious in the intervention of airway remodeling in COPD at different time periods.
