Background:Coronary heart disease as the most common clinical cardiovascular disease, is one of the highest human fatality rate of chronic diseases. Percutaneous coronary angioplasty surgery (PTCA) and coronary artery bypass graft (CABG) the treatment of rapid development, and has been widely promoted, but at the same time there are problems:myocardial ischemia-reperfusion injury seriously affect the clinical efficacy of coronary heart disease after surgical treatment, we need to focus, to overcome its effects. Salvia traditional Chinese medicine as blood circulation, commonly used drug for coronary heart disease treatment, extract of Salvia miltiorrhiza water-soluble active ingredients salvianolic acids by intravenous administration, validation of salvianolic acids on ischemia and reperfusion after myocardial protection. Studies have shown that salvianolic acids in the use of Salvia miltiorrhiza inhibit the inflammatory factors reduce oxygen free radicals, to protect the myocardium, through the experimental analysis of salvianolic acids activate PKC-epsilon in cardiac myocytes, expression of HSP70after the intervention in myocardial ischemia and reperfusion the molecular mechanisms. By analyzing the molecular mechanism of salvianolic acids protect the myocardium, to clear the way of its role, provide a clear prescription basis for clinicalObjective:1,To understand the pathogenesis of myocardial ischemia and reperfusion injury, salvianolic acids intravenous explained by biochemical, pathological, and other indicators of ischemia and reperfusion injury after myocardial protection;2, the molecular mechanism investigate the myocardial protective effect of salvianolic acids, and further analysis of salvianolic acids activate the myocardial cells of PKC-epsilon, the molecular mechanisms of HSP70expression after the intervention in myocardial ischemia and reperfusion, and provide a clear prescription basis for clinical.Materials and Methods:80healthy adult wistar rats were randomized to the normal group, model group, treatment group and blocker group, sham operation group (n=10)15. In addition to the normal group and sham group, the rest of the implementation of in vivo myocardial ischemia and reperfusion surgery, ligating the left coronary artery,30min after releasing the ligature. The experimental data SPSS16.0statistical package for statistical analysis.Results:1,Cardiac function and morphology results:Compared with model group, treatment group rats after reperfusion LVSP increased (P<0.05), LVDP decreased (P<0.05),+dp/dt max increased (P<0.05),-dp/dtmax lower (P<0.05) after reperfusion, the heart weight index decreased (P<0.05), the ratio of myocardial infarction area narrowing (P<0.05), the light and electron microscopy of morphological changes in model group, but more important than the sham operation group, cTn-T, clear and TNF-a levels decreased significantly (p<0.05), serum and myocardial tissue MDA significantly decreased the activities of SOD, T-AOC was significantly increased, the difference was statisticallysignificance (P<0.05or P <0.01), the indicators were statistically significant.2, the molecular mechanism results:Compared with model group of PKC-epsilon in the mitochondria of the treatment group were significantly increased, while the cytosolic PKC-epsilon content was significantly decreased, the intracellular content of HSP70was significantly increased, the three indicators both differences statistically significant (P <0.05).Conclusion:1.Salvianolic acids regulate arrhythmia to a certain extent, improve heart function and reduce myocardial ischemia and reperfusion live injury infarct size, improve the degree of myocardial damage, the maintenance of myocardial tissue structure and morphology, an increase of antioxidant enzymes, reducing the the generation of oxygen free radicals, reduce the inflammatory response, in order to protect myocardial cells against myocardial reperfusion injury.2,of salvianolic acids activate the myocardial cells after ischemia and reperfusion of PKC-epsilon, and the role of the mitochondria, while increasing the intracellular expression of HSP7O, in order to protect myocardial cells.
