The Experimental Influences Of Baihui-qubin On AIF Expression Of Rats With Acute Intracerebral Hemorrhage

Objective:Throuth the therapeutic process of acupuncturing Baihui-Qubin,observe the changes of the expression of apoptosis inducing factor(AIF protein)during acupuncture treatment,so as to explore its effect of the expression of AIF in the brain tissue of rats with acute intracerebral hemorrhage,and to provide a favorable basis for the treatment of acute intracerebral hemorrhage.Methods:75 male Wistar rats were randomly divided into blank group,model group and acupuncture group.Among them,there were 15 rats in blank group,30 rats in model group and acupuncture group respectively.Each group was equally divided into five points:6h,12h,24h,3d,and 7d.Build the rat model of cerebral hemorrhage by autologous blood injection in model group and acupuncture group.When the rats revived,observe and record the degree of neurological impairment,then select the qualified model.The acupuncture group use acupuncture treatment of Baihui-Qubin.The rats in each group were observed systematically:(1)To observe the score of neural function defect at different time points in each group of rats;(2)The condition of apoptosis around hematoma after intracerebral hemorrhage in each group of rats was detected by TUNNEL;(3)Immunohistochemical method was used to detect the dynamic expression of AIF in the hematoma around intracerebral hemorrhage.Results:(1)The model group and the acupuncture group began to show signs of neural function defect at 6h;At 24h,the signs of neurological impairment was very significant;when 3th day,the signs were severest,until the 7th day,there was a little signs of neurological impairment still exist.The neural function score of acupuncture group was significantly improved than model group when compared at the same time point,and there were significant differences in 12h,24d and 3d(P<0.01);(2)The results of TUNNEL showed that compared with model group,the number of nerve cell apoptosis in acupuncture group decreased in different degrees at each time point,and there was a significant difference in 24h,3d and 7d(P<0.01);(3)Immunohistochemical detection result showed that compared with the model group,the level of AIF protein in the acupuncture group decreased at each time point,and there was a significant difference in 24h,3dand 7d(P<0.01)Conclusions:(1)The acupuncture treatment of Baihui-Qubin can reduce the symptoms of nerve function defect of cerebral hemorrhage in rats;(2)The acupuncture treatment of Baihui-Qubin can effectively suppress the apoptosis of neurons after ICH;(3)By inhibiting the expression of AIF,the acupuncture treatment of Baihui-Qubin can protect the brain tissue around the hematoma after intracerebral hemorrhage.
