Inquiring Into The Mechanism Of Kuntai Capsule Treating Decreasing Ovarian Reserve By The Regulation Of Granular Cell Apoptosis

Objective:Sprague-Dawley female rats model of DOR was established by intraperitoneal injection of VCD of 80mg/kg continuously 15 days.To study the effects of Kuntai Capsule on the general conditions of the DOR rats,Follicle-stimulating(FSH),lactogenichormone(LH),estradiol(E2),the expression of Bcl-2,Bcl-xl,Bax and Bad protein,the HE of the ovaries and uteruses and the condition of offspring of the DOR rats.It offers theoretical foundation to prevent and treat DOR by traditional Chinese medicineMethod:The model of DOR was established by intraperitoneal injection of VCD of 80mg/kg continuously 15 days.At the beginning of modeling.intragastric administration had started and lasted 45 days.They were divided into the black control group,the model control group,the high,middle and low dosage Kuntai Capsule,and the positive control medicine of Estradio Valerate.To study the pregnancy,still birth rate and the condition of offspring.FSH,LH and E2 were detected by ELISA method.To observe the histological change of the ovaries and uteruses by HE.The Expression levels of Bcl-2.Bcl-xl Bax and Bad protein in rats ovarian were detected by Western-blot method.Results:1、Compared to the black control group,the rats of model control group were slow in reacting;compared to the model control group,after medication the rats were quick in reacting.And their hair colour changed better.2、The model control group were the skinniest in comparison with the black control group.They grow slowly.lt possessed extreme difference(P<0.01):After intragastric administration the rats improved inordinately.Compared to the model control group,the low dosage Kuntai Capsule group weight increased(P<0.05).The middle.high dosage Kuntai Capsule group and Estradio Valerate group increased outstandingly(P<0.01).3、By pathological examination,The ovaries and utenuses were shrinked remarkablely.The amount of the primordial follicles and primary follicles reduced obviously.The endometrium were thin.After taking the medication,They got different improvement.4.Compared to the black control group,the FSH of the model control group increased strikingly,E2 decreased(P<0.05).LH changed unconspicuous.Compared to the model control group,The FSH of the high,middle and low dosage Kuntai Capsule group decreased,E2 increased(P<0.05).LH improved unapparently.FSH、LH of positive control medicine of Estradio Valerate group decreased and E2 increased(P<0.05).5.The expression of Bcl-2 and Bcl-xl protein of model group decreased obviously and the the expression of Bax and Bad protein of model group increased(P<0.05),Bcl-2/Bax ratio of model group decreased sharply(P<0.01).Compared to the model control group,the expression of Bcl-2 and Bcl-xl protein of high-dosage Kuntai Capsule group and Estradio Valerate group increased obviously(P<0.01-0.05);the expression of Bcl-2 and Bcl-xl protein of middle and low dosage Kuntai Capsule group increased(P<0.05),the expression of Bax and Bad protein of different dosage group and Estradio Valerate group decreased(P<0.05).Bcl-2/Bax ratio of middle and low dosage Kuntai Capsule group increased(P<0.05),and the ratio of high-dosage Kuntai Capsule group and Estradio Valerate group increased evidently(P<0.01).6.There was no statistic difference in the pregnancy rate of each goup;the model control groups still birth rate was 14%,the low dosage Kuntai Capsule group was 3%.There was no still birth rate in the remaining group.7.VCD produced blight to the offspring.Compared to the black control group,the offspring of the model control group were lean.After having the treatment,the middle,high-dosage Kuntai Capsule group and Estradio Valerate group got improvement,but the low-dosage Kuntai Capsule group changed non-significantly.Conclusion:1.Kuntai Capsule can improve the general condition of DOR rats.2.Kuntai Capsule can improve themorphology of ovaries and uteruses.3.Kuntai Capsule can lower FSH、LH and up-regulate E2.It can control granule cells apoptosis,putting off senium.4.Kuntai Capsule can improve the function of ovaries by up-regulating the expression of Bcl-2 and Bcl-xl protein and down-regulating the expression of Bax and Bad protein in ovarian.5.VCD produced blight to the offspring,on the contrary Kuntai Capsule can protect the offspring.
