Objective:Our study aimed to assess the association of the gene expression levels in mRNA of IRAK4 and IRF5 with ischemic stroke?IS?of wind phlegm stasis syndrome and its related quantitative traits.We hope that the study will further elucidate the biological basis of IS of wind phlegm stasis syndrome,providing a genetic and biological basis for revealing the scientific connotation of TCM syndrome in it.Methods:The case-control study including 34 IS of wind phlegm stasis syndrome patients and 57 controls were investigated in First Affiliated Hospital of Guangxi University of Chinese Medicine during June 2014 to December 2016.The former were treated in Department of Internal Neurology and latter were participated in physical examination or patients without nervous system diseases and serious illness.Collecting information about cases and controls,such as name,age,ethnic,gender and so on.Diseases in the past time,such as hypertension,diabetes,high blood lipids and so on.Habits about smoking,drinking and so on.Genetic disease and so on.Chief complaint,process of the disease,evidence of the disease,diagnosis and diagnosis of TCM.Detecting Blood pressure,total triglyceride?TG?,cholesterol?TC?,high density lipoprotein cholesterol?HDL-C?and low-density lipoprotein cholesterol?LDL-C?.Peripheral blood samples were used to detect IRAK4 and IRF5 gene expression levels by real-time PCR technology.Statistical analyses were carried out with SPSS16.0 for windows.Results:1.Association of the gene expression levels of IRAK4 and IRF5 with IS of wind phlegm stasis syndrome:the expression levels of IRAK4 gene in IS of wind phlegm stasis syndrome group were significantly reduced?P=0.017?.The expression levels of IRF5 gene in IS of wind phlegm stasis syndrome group were significantly increased?P=0.017?.2.Association of the gene expression levels of IRAK4 and IRF5 with related quantitative traits about IS of wind phlegm stasis syndrome:No statistically significant association were found between the expression levels of IRAK4 gene and normal range and abnormal range about SBP,DBP,blood sugar,TC,TG,HDL-C and LDL-C in cases(Padj>0.050).No statistically significant association were found between the expression levels of IRF5 gene and normal range and abnormal range about SBP,DBP,blood sugar,TC,TG and LDL-C in cases(Padj>0.050).Statistically significant association were found between the expression levels of IRF5 gene and normal range and abnormal range about HDL-C in cases(Padj<0.050).3.Calculate with Pearson correlation coefficient about association of the gene expression levels of IRAK4 and IRF5 with related quantitative traits about IS of wind phlegm stasis syndrome:No statistically significant association were found between the expression levels of IRAK4 and IRF5gene and normal range and abnormal range about SBP,DBP,TC,TG,HDL-C and LDL-C in cases(Padj>0.050).Statistically significant association were found between the expression levels of IRF5 gene and normal range and abnormal range about blood sugar in cases(Padj<0.050),but not in IRAK4(Padj>0.050).Conclusion:1.The gene expression levels of IRAK4 may be related to IS of wind phlegm stasis syndrome.2.The gene expression levels of IRF5 may be related to IS of wind phlegm stasis syndrome.3.The gene expression levels of IRAK4 may not influence the levels about SBP,DBP,blood sugar,TC,TG,HDL-C and LDL-C in IS of wind phlegm stasis syndrome.The gene expression levels of IRF5 may not influence the levels about SBP,DBP,blood sugar,TC,TG and LDL-C in IS of wind phlegm stasis syndrome.The gene expression levels of IRF5 may influence the level about HDL-C in IS of wind phlegm stasis syndrome.4.The gene expression levels of IRAK4 may be not related to the levels about SBP,DBP,blood sugar,TC,TG,HDL-C and LDL-C in IS of wind phlegm stasis syndrome.The gene expression levels of IRF5 may be not related to the levels about SBP,DBP,TC,TG,HDL-C and LDL-C in IS of wind phlegm stasis syndrome.It may be positive correlation between the gene expression levels of IRF5 and the level about blood sugar in IS of wind phlegm stasis syndrome,which may be reference indicator.
