Objective:With the improvement of people's living conditions,many unusual diseases such as high blood pressure,diabetes incidence increased year by year,and changes in living environment also led to an increase in infertility patients.Although the emergence of assisted reproductive technology met the fertility needs of some patients,but the success rate of pregnancy still didn't reach the desired level.In vitro fertilization-freeze-thaw embryo transfer process,the success factor in addition to excellent embryonic quality,also depends on endometrial receptivity.It is known that pinopodes are expressed on the endometrium during the embryo implantation window,and pinopode measurement has been proposed as a marker of endometrial receptivity.By detecting the pinopode expression during the implantation window and analyzing the outcome of the pregnancy after embryo transfer,our research group confirmed that pinopodes can be used as a marker of endometrial receptivity,pinopode measurement has a prognostic value of successful pregnancy after embryo transfer.Methods:172 infertile women undergoing frozen embryo transfer following IVF treatment were recruited.Among 172 subjects,46 women took part in the first study to quantify the daily changing pattern of pinopodes 3 to 7 days after the imitation of progesterone therapy in the hormone replacement cycles.The remaining 126 cases participated in a study to examine the relationship between pinopode count and pregnancy following frozen embryo transfer in hormone replacement cycles.Scanning electron microscopy was used to capture the pinopode image,followed by using imageJ program to quantify the count and subtype of the pinopodes.Estimate the minimum number of fields required for the assessment and use an improved pinopode scoring method for quantitative analysis.Results:(1)At least 60 fields were necessary to produce a reproducible result.(2)The total pinopode count reached a peak on day P+6,and then dropped slightly.The mean number of fully developed pinopode per field count also reached a maximum on day P+6.(3)The expression of pinopode was significantly different between subjects who did and did not conceive.Conclusions:The examination of the number and morphology of pinopodes is of great help to the evaluation of endometrial receptivity,which has a high prognostic value for successful pregnancy after embryo transfer.
