The Study On The Contraction Effect And Mechanism Of Several Traditional Chinese Medicine Prescription On Isolated Varicose Veins

ObjectiveThe pathogenesis of lower limb chronic venous insufficiency is not yet clear.The theory of vascular remodeling is the key of CVI and the hotspot in recent years.CVI patients with lower extremity venous hypertension,venous valve insufficiency,blood reflux disorders,chronic inflammation,the formation of varicose veins,calf soreness,limb edema,pigmentation,lipid sclerosis and ulcer clinical symptoms.There has a series of changes in the venous wall composition,structure and function of CVI patients.The change make up with vascular remodeling,resulting in decreased flexibility of the venous wall,systolic dysfunction,tension weakened.In the process of CVI progression,venous hypertension,venous valve insufficiency,reflux disorders and other pathogenic factors common pathology showed a decrease in venous wall tension.It can be seen that the weakening of venous tension plays an important role in the course of CVI.Traditional Chinese medicine holds that CVI belong to "jin liu"?"lian chuang"?"e mai" category,the type of evidence mainly liver and qi stagnation syndrome,spleen deficiency dampness syndrome,fatigue,blood stasis syndrome.Chinese medicine treat different CVI has clear effect and little side effects,it is worth promoting.Combined with clinical practice,according to dialectical classification were selected Chai Hu Shu Gan San,Yu Ping Feng San,Shen Ling Bai Zhu San,Tao Hong Si Wu Tang.The four Chinese medicine as an experimental drug to addition on the vascular venes,to observe the effect on the tension of the venes wall and the contraction of the vein in vitro,trying to discussion mechanism,and the Diosmin as a Western medicine control.In this study,we investigated the relationship between Chinese herbal medicine and venous tension by observing the contraction effect vitro vein,and provided the laboratory basis for the treatment of CVI,provided new ideas for the development of new drugs.MethodsWe used isolated varicose veins of the lower limbs and allantoic veins as the experimental object,recorded the effects of tension on the isolated venous ring using several Chinese herbal compounds and diosimin by microcirculation tension tester respectively,recorded the Chai Hu Shu Gan San's effects on isolated varicose vein after incubated with nifedipine.SPSS 19.0 statistical software was used to analyze the data.Statistical analysis was carried out by one-way ANOVA and Covariance analysis.The concentration-response curve was fitted by four-parameter logistic model,and the regression equation for F test,the regression parameters into t test;the curve using Origin 9 mapping and analysis.Results1.Four traditional Chinese medicine compound can shrink the varicose veins in a concentration-dependent manner,of which Shen Ling Bai Zhu San contraction capacity of the strongest,followed by Chai Hu Shu Gan San and Yu Ping Feng San,Tao Hong Si Wu Tang weakest.2.Diosmin have no obvious contraction on the varicose veins.3.Nifedipine can significantly inhibit the contraction of Chai Hu Shu Gan San on varicose veins.ConclusionChai Hu Shu Gan San,Shen Ling Bai Zhu San,Yu Ping Feng San and Tao Hong Si Wu Tang can shrink the varicose veins in vitro;Chaihu Shugan Powder on the contraction of varicose veins may be related to the promotion of extracellular calcium influx.Diosmin has no direct contraction of the venous vein,and its role in improving the venous tension may be achieved through other mechanisms of action.
