Objective â‘ To observe the microstructural and ultrastructural changes of the varicose great saphenous vein. â‘¡To observe the distributing and changes of elastic fiber (EF) and collagenous fiber (CF) in the varicose great saphenous vein. â‘¢To observe the expressive changes of Cathepsin (Cat) L, S, K, B and Cystatin C in the varicose great saphenous vein. â‘£To investigate the relationship between the varicose great saphenous vein and the Cat L, Cat S, Cat K, Cat B, CystatinC. Methods 31 samples of varicose great saphenous vein and the 23 samples of normal great saphenous vein were obtained from operation, and ultrastructural changes of the veins were observed by transmission electron microscope. The histological changes were observed by hematoxylin eosin and Weigert staining. The expressive changes of Cat L, Cat S, Cat K, Cat B and Cystatin C in the veins were studied by immunohistochemical SABC. Colocalization of Cat L, Cat S, Cat K, Cat B and Cystatin C with cell-types in the veins were studied by immunofluorescent colocalization. The optic density of Cat L, Cat S, Cat K, Cat B and CystatinC immunoreactive was analyzed by computerized image pattern analysis, all experimental results were statistically analyzed with SPSS 12.0. Results â‘ In HE staining sections, normal great saphenous vein whose cavity smooth were characterized by thin tunica intima. The tunica media mainly consisted of long spindle-shaped smooth muscle cell (SMC) which ring-like layered arrangement. The tunica adventitia was thicker. Varicose great saphenous vein demonstrated the diameter increased. The tunica intima accumulated and the number of SMC was increased.
