Purpose The effect of Traditional Korean medicine Mahuangfabiao Decoction on airway inflammation in asthmatic mice.Methods 40 BALB/c female mice were randomly divided into 5 groups:normal control group,asthma model group,traditional Korean medicine Mahuangfabiao decoction high dose group,traditional Korean medicine Mahuangfabiao decoction low dose group,dexamethasone positive control group.Except for the normal control group,the rest of the four group mice were sensitized on the 1st,7th and 14th day with a mixture of 100?g of protein and 4mg of aluminum hydroxide powder,the normal control group was injected intraperitoneally with 0.9%sodium chlorid.On the 21st day,the mice except the normal control group were given 3%OVA inhalation in 4 days,and the normal control group was inhaled with 0.9%sodium chloride under the same conditions.Mahuangfabiao decoction high dose group and low dose group and dexamethasone positive control group were given gavage for 1 week from day 18.Normal control group and asthmatic model group were treated with 0.9%sodium chloride.Mice lung tissue and bronchoalveolar lavage fluid were extracted 1 day after the last challenge,the pathological changes of lung tissue were observed by HE and PAS staining,Diff-Quick was used to observe the type and count of inflammatory cells in BALF,IL-4,IL-5,IL-13 and IFN-? were measured by ELISA,effects of Mahuangfabiao decoction on the expression of NF-?B p65 and p-p38MAPK in lung tissue of asthmatic mice were measured by immunohistochemical staining and immunoblotting.Results 1.Compared with the normal control group,the alveolar and bronchial wall of the asthmatic model group were destroyed,the luminal secretions increased,,the airway epithelial cells were destroyed,the EOS gathered in large numbers,and the airway mucosa showed a lot of goblet cell hyperplasi by observing the lung tissue using HE and PAS staining.Mahuangfabiao decoction high and low dose group compared with the model group,the degree of lung injury is light.2.Diff-Quick staining was used to observe and count the inflammatory cell types in BALF and the counting results show that asthma model group see a large number of aggregated EOS,the total number of cells and three cell counts,including EOS,NE,lymphocytes increased significantly,compared with the normal control group were significantly different(P<0.05)Mahuangfabiao decoction high and low dose group compared with the model group,there is a small amount of aggregation of EOS and the total number of cells,three cell counts were significantly reduced(P<0.05).3.The levels of IL-4,IL-5 and IL-13 in BALF of asthmatic mice model group were significantly increased,IFN-y was significantly decreased by ELISA compared with normal control group(P<0.05)The levels of IL-4,IL-5 and IL-13 in BALF of Mahuangfabiao decoction high and low dose group were decreased and the content of IFN-? was increased.Compared with asthmatic model group,there were significant differences(P<0.05).4.Immunohistochemical staining was used to observe the expression of NF-?Bp65 in the lung tissue of each group.The results showed that the nuclei of asthmatic model group were stained with dark brown,compared with the normal control group,the expression of NF-?Bp65 protein was significantly increased.Compared with asthmatic model group,there is a small amount of dark brown yellow nucleus around the bronchus in the Mahuangfabiao decoction high and low dose group and dexamethasone positive control group,and the NF-?Bp65 protein expression was significantly decreased.5.The content of p-p38MAPK protein was detected by Western blotting,the results showed that p-p38MAPK expression in asthmatic mouse model group was significantly higher than that in normal control group.The expression of p-p38MAPK in the Mahuangfabiao decoction high and low dose groups were lower than that in the asthmatic model group.Conclusion 1.Mahuangfabiao decoction can relieve airway inflammation in bronchial asthma mice.2.Mahuangfabiao decoction can downregulate levels of inflammatory cytokines IL-4,IL-5 and IL-13,upregulate IFN-y levels,thereby regulating the balance of Thl/Th2 and inhibiting airway inflammation in asthmatic mice,the mechanism may be through down-regulation of NF-?Bp65,p-p38MAPK expression.
